肥胖是一種全身系統性慢性發炎並且具有高氧化壓力,為許多疾病的危險因子。研究指出芥子酸具抗氧化、抗發炎及神經保護性等效果。但是,芥子酸是否可影響肥胖所引起之發炎及氧化壓力等相關研究甚少,因此,本實驗欲探討芥子酸是否能於高脂飲食誘發肥胖之模式下,降低小鼠氧化壓力及發炎反應,以減緩因肥胖所引發的胰島素阻抗、脂肪堆積等。本實驗動物採用雄性C57BL/6JNarl小鼠,餵食高脂飼料2週以誘發小鼠肥胖,再以體重為依據做S型分組,分為:(1)正常組、(2)高脂飲食組、(3)高脂飲食+低劑量芥子酸組(0.5 mg芥子酸/隻/天)、(4)高脂飲食+高劑量芥子酸組(1.0 mg芥子酸/隻/天)。本實驗共經7週後犧牲,採集血液、心臟、肝臟、腎臟、脾臟、副睪脂肪進行樣本分析。實驗結果顯示,高油飲食組有較高胰島素分泌抗性指數(HOMA-IR),並且顯著增加體重、副睪脂肪重、肝臟三酸甘油酯以及前發炎因子如,細胞介素-6、腫瘤壞死因子-α。在肝臟氧化壓力部分,榖胱甘肽濃度與榖胱甘肽過氧化酶酵素活性下降、氧化型榖胱甘肽蛋白濃度上升。給予芥子酸後,顯著降低小鼠體重、副睪脂肪重量、肝臟三酸甘油酯濃度、提高榖胱甘肽濃度與榖胱甘肽過氧化酶酵素活性以及顯著降低氧化型榖胱甘肽濃度。研究結果推測芥子酸可能經由其抗氧化及抗發炎的效果改善因高脂飲食引起的併發症。

Obesity is associated with a complex systemic inflammatory and oxidative state. It has been considered as a crucial risk factor responsible for the development and progression of many diseases. Several studies about sinapic acid (SA) have shown that it possesses anti-oxidative, anti-inflammatory and peroxynitrite scavenging effects. However, it is known that SA could attenuate inflammatory and oxidative stress in obesity. In this present study, the protective effects of SA on high fat diet induced obesity mice were examined. This study had four groups. C57BL/6JNarl male mice were fed by high-fat diet for two weeks and divided into three groups: (1) high fat diet (HF) group ; (2) HF+low dose SA group (HF+LS) (0.5mg SA/mouse/day) ; (3) HF+high dose SA group (HF+HS) (1.0mg SA/mouse/day). Normal group were fed with normal diet in whole experimental period. At the end of the seventh week, mice were killed and the tissues were collected for analyses. Results showed HF group had significantly higher body weight, epididymal adipose tissue and liver triglyceride level than normal mice. However, mice with SA intake had significantly lower body weight, epididymal adipose tissue and liver triglyceride level. Furthermore, SA intake retained liver GPx activity and GSH content. These findings support that sinapic acid through its anti-oxidative and anti-inflammatory effects could attenuate the complications caused by high-fat diet.
