馬藍(Baphicacanthus cusia (Nees) Bremek.)為爵床科(Acanthaceae)植物,整株植物皆入藥,其根部稱板藍根(南板藍根),葉部稱大青葉,而其葉或莖葉經加工後製成的乾燥粉末或團塊稱為青黛。依文獻記載馬藍為板藍根、大青葉及青黛的基原植物之一,具有抗發炎、抗菌及抗癌等活性,馬藍根部之抽出物在細胞毒性方面對p-388具有很強的活性,因此本研究以細胞毒性當活性引導分離指標,進行成分分離。
馬藍的乾燥根之甲醇抽出物經分離及結構鑑定,分出成分為羽扇醇lupeol (H-1), stigmasterol和b-sitosterol混合物 (H-2), 樺木醇 betulin (H-3), 靛藍indigo (C-1), 靛玉紅indirubin (C-2), 2-benzoxazolinone (E-1)及2-hydroxy-1,4-benzoxazin-3-one (E-2), 香草酸 4-hydroxy-3-methoxy benzoic acid (E-3)。其中羽扇醇lupeol (H-1), stigmasterol和b-sitosterol混合物 (H-2), 樺木醇betulin (H-3), 靛藍indigo (C-1)及靛玉紅indirubin (C-2)等為主成分,而2-benzoxazolinone (E-1), 2-hydroxy-1,4-benzoxazin-3-one (E-2), 4-hydroxy-3-methoxy benzoic acid (E-3)係首次由本植物中分離得到。

The plant Baphicacanthus cusia (Nees) Bremek. of Acanthaceae family has been used in traditional Chinese medicine for a long time. The roots were taken as Ban-Lan-Gen (Baphicacanthus Radix and Rhizoma), the leaves were used as Ta-Chin-Yeh (Baphicacanthus Folium) while Chin-Dai was prepared from the leaves of Baphicacanthus cusia. The plant has been used as anti-inflammatory, antibacterial and anti-carcinogenic agent. The extract of Baphicacanthus cusia root showed strong inhibitory effect on tumor cell lines such as p-388. Based on the cytotoxicity guided chemical fractionation, the bioactive compounds would be isolated and the chemical structures be elucidated.
The methanolic extract of the root was partitioned and separated by chromatographic methods. The eight compounds were isolated and identified. Their structures were determined on the basis of spectral evidence as well as direct comparison with authenic samples. These components included lupeol (H-1), mixture of stigmasterlol and β-sitosterol (H-2), betulin (H-3), indigo (C-1), indirubin (C-2), 2-benzoxazolinone (E-1), 2-hydroxy-1,4-benzoxazin-3-one (E-2), 4-hydroxy-3-methoxy benzoic acid (E-3). Among these , 2-benzoxazolinone (E-1), 2-hydroxy-1,4-benzoxazin-3-one (E-2), 4-hydroxy-3-methoxy benzoic acid (E-3) were isolated for the first time from this plant.
