甜椒疫病菌Phytophthora capsici之粒線體DNA經由三個RAPD引子增幅後，選取三個增幅片段與載體PCR-ScriptTM Amp SK(+)結合，再轉型至E. coli CL 1菌株，得到三個選殖株(clones)，分別命名為pPCA90、pPCA85、pPCA80，其插入片段大小分別為900 bp、850 bp及800 bp。這些選殖株以非放射性之Fluorescein Gebe Images TM Labelling (Amersham, Buckingamshire, England)標示作為核酸探針，並以南方轉漬法(Southern blotting)及點漬法(Dot blotting)分析台灣不同的區域所採集的菌株，露菌病等全量DNA之雜合關係。初步分析結果，pPCA80較具有專一性。

There specific RAPD products of amplified Mitochondiral DNA fregments of Phytophthora capsici were cloned by the vector pCR-Script TM Amp SK(+). We have developed 3 probes called pPCA90, pPCA85 and pPCA80 according to their insert size are 900bp, 850bp and 800bp. Labelled by non-radiation Fluorescein Gene Images TML abelling kit, There were Tested specifictivity by southern blot and dot blot with P. capsici and Perophythora litchii isolates from Taiwan. The primary sesult is pPCA90 more specific to P. capsici. It will be testing specifictivity more with more isolates.
