脂質代謝是維持正常生理功能及生命所必須。然而在已開發或開發中國家,脂質代謝的失調,或稱脂質代謝異常卻逐漸成為一個嚴重的健康問題。高血脂症:諸如高三酸甘油脂或是高膽固醇是最常見的一種脂質代謝異常並且常伴隨著肥胖或是粥狀動脈硬化的發生。近年來,臨床上對於這類疾病的用藥治療已經轉移到改善與其相關的脂質代謝異常上。而在最近幾年,中藥在預防人類疾病的使用上也逐漸受到關注。漢黃芩素是中藥黃芩的主要萃取物之一,並且已經被報導具有許多不同的抗發炎及抗氧化的功效。然而,漢黃芩素對於脂肪細胞中三酸甘油脂的代謝及巨噬細胞中膽固醇的代謝的影響至今仍未十分明瞭。因此在本篇研究中,我們主要是針對漢黃芩素對於這兩個脂質代謝途徑的探討及其可能參與的機制。我們發現漢黃芩素並不會改變3T3-L1前脂肪細胞的生長及分化。然而,在成熟的脂肪細胞中,漢黃芩素會透過蛋白質激?,來刺激脂質分解的速率。而在正常老鼠中,漢黃芩素同樣會影響脂質的代謝,包括減少白色脂肪組織、增加高密度脂蛋白的膽固醇含量、降低血清中的三酸甘油脂、改變血中脂肪激素的含量、抑制脂肪生成相關蛋白的表現和促進脂質分解作用。由此結果可以得知,漢黃芩素在活體中不但具有調節脂質生成和脂質分解作用,並具有預防高血脂症的效果。在粥狀動脈硬化病變的形成中,巨噬細胞內會大量堆積膽固醇並形成泡沫細胞且調控膽固醇的代謝。而我們發現在巨噬細胞中,漢黃芩素的給予可以抑制氧化態低密度脂蛋白所造成的脂質堆積,且主要是透過了增加三磷酸腺?ㄤ畢X盒轉運體A1所驅使的膽固醇流出而達到此保護作用。我們也證實,蛋白質激?,和外源鈣/鈣調素依賴型激?〞滌T息傳遞路徑在漢黃芩素抑制泡沫細胞的形成和增加三磷酸腺?ㄤ畢X盒轉運體A1的表現等作用上扮演關鍵的角色。此外,漢黃芩素增加三磷酸腺?ㄤ畢X盒轉運體A1的蛋白表現量主要是增加了其蛋白質的穩定性,而非影響到其基因的轉錄。藉由使用免疫共沉澱的實驗方法及蛋白質去磷酸??B的抑制劑,我們也發現漢黃芩素延長三磷酸腺?ㄤ畢X盒轉運體A1的半衰期是經由增加其與蛋白質磷酸??B的結合而導致被去磷酸化所達到的,此結果也進而減緩了細胞內脂質的堆積。由此結果我們可以得知,漢黃芩素可以透過三磷酸腺?ㄤ畢X盒轉運體A1將膽固醇送出,而抑制泡沫細胞的形成。總的來說,本篇研究證實了漢黃芩素具有預防肥胖及治療粥狀動脈硬化的潛力。
Lipid metabolism is essential for maintenance of physiological function and life. However, dysregulation of lipid metabolism, also called lipid abnormality, has been considered as a severe health problem in developing or developed countries. Hyperlipidemia, including higher level of serum triglyceride or cholesterol, is the most common lipid abnormality and associates with the obesity and atherosclerosis. Over the past decade, the ultimate goal of clinical trails for therapy of obesity or atherosclerosis has been shifted to explore the new classes of agents, which also can provide protection against lipid abnormality. Recently, Chinese herbs have received increasing attention and been extensively used for the prevention of human diseases. Wogonin, the Scutellaria baicalensis Georgi extracts, have been reported to exert protective effects on inflammation and oxidative stress. However, effects of wogonin on triglyceride metabolism in adipocytes and cholesterol metabolism in macrophages are not fully understood. In the present study, we would like to investigate the effect of wogonin on these two processes. We examined that treatment of wogonin does not affect the proliferation and differentiation in 3T3-L1 preadipocytes. Additionally, wogonin administration significantly increased the adipocyte lipolysis through the protein kinase A (PKA)-dependent mechanism. The effects of wogonin on lipid metabolism, including decreased white adipose tissue mass, elevated high-density lipoprotein-cholesterol (HDL-c), reduced serum triglyceride, altered adipokine secretion and adipogenesis-related protein and enhanced lipolysis, were also defined in WT mice. These results suggest that wogonin modulates adipogenesis and lipolysis and preserves the preventive effects on hyperlipidemia. During atherogenesis, foam cells, which are characterized by the over-loaded cellular cholesterol in macrophages, play the critical role in cholesterol metabolism. Here, we showed that wogonin attenuated oxidized low-density lipoprotein (oxLDL)-induced cholesterol accumulation in macrophages. The suppressive effect of wogonin was due to the increase of ABCA1-dependent cholesterol efflux. Besides, activation of PKA and calcium/calmodulin-dependent protein kinase is necessary for the inhibitory effect of wogonin on formation of foam cell. Moreover, the up-regulation of ABCA1 protein by wogonin resulted from a decrease in degradation rate of ABCA1 protein, whereas wogonin did not affect the mRNA expression of ABCA1. We show that wogonin extended ABCA1 half-life via protein phosphatase 2B (PP2B)-mediated dephosphorylation, as evidenced by an increase in interaction between ABCA1 and PP2B. Otherwise, pharmacological inhibition of PP2B prevented wogonin-induced ABCA1 protein expression, dephosphorylation and attenuation of lipid accumulation. These results indicate that wogonin sufficiently inhibits the formation of macrophage foam cells via ABCA1-dependent cholesterol transport. Collectively, our study provides the potency of wogonin in the regulation of peripheral triglyceride and cholesterol metabolism.
