乙醯胺酚 (Acetaminophen, APAP)是一種廣泛使用的止痛和退熱的藥物,當APAP過量時易造成急性肝臟中毒。魚腥草 (Houttuynia cordata Thunb)為三白草科蕺菜屬植物。魚腥草搓碎有魚腥味,口感略澀,乾燥後可以去除魚腥味。民間認為魚腥草可以清熱解毒,排膿消腫,利尿通淋。本研究欲探討魚腥草對APAP誘發小鼠急性肝損傷是否具有保護效果。將30隻C57BL/6JNarL小鼠隨機分為正常組、控制組、8%魚腥草組共三組。飲用5週後,腹腔注射APAP並於24小時後犧牲,收集血液和臟器組織,進行肝功能、抗氧化和抗發炎等分析。結果顯示,給予魚腥草水煮液能顯著改善APAP所造成之肝功能指數 (AST、ALT)上升;減緩抗氧化活性 (GSH、GPx、SOD、CAT、TAC)下降;降低發炎相關因子 (IL-6、TNF-α、MCP-1)的釋放;以及可抑制肝臟CYP2E1活性和脂質過氧化MDA之產生。實驗結果支持魚腥草水煮液可以保護肝臟抗拒APAP誘發小鼠的急性肝損傷。

Acetaminophen (APAP), a commonly used antipyretic and analgesic agent, could cause acute liver injury by overdose. Houttuynia cordata Thunb is a medicinal plant used in traditional Chinese medicine. The aim of this study was to examine the hepatoprotective effects of Houttuynia cordata Thunb aqueous extract (HCE) against APAP-induced injury. Male C57BL/6JNarL mice were randomly divided into normal group, control group and HCE group. After 5 weeks supplementation, mice were treated by APAP intraperitoneally, and killed after 24 hours. Blood and organs from each mouse were collected. Results showed that the intake of HCE significantly alleviated APAP-induced aspartate aminotransferase (AST) and alanine aminotransferase (ALT) elevation; retained glutathione (GSH), glutathione peroxidase (GPx), superoxide dismutase (SOD), catalase (CAT) and total antioxidant capacity (TAC) activities or levels; reduced the level of inflammatory related factors such as interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α) and monocyte chemotactic protein-1 (MCP-1). Furthermore, HCE intake attenuated cytochrome P2E1 (CYP2E1) activity and malondialdehyde (MDA) production. These findings supported that pretreatment of HCE protected mice liver against APAP-induced oxidative and inflammatory injury.
