海獸胃線蟲(Anisakis simplex)係存在於海中魚類及海洋哺乳類,人類食用海獸胃線蟲感染的海產後會引起人類海獸胃線蟲過敏症。本工作對白帶魚(Trichiurus lepturus)、白腹鯖(Scomber japonicus) 、真��(Trachurus japonicus)等三種魚體內之海獸胃線蟲進行調查,期望了解台灣基隆嶼海域的感染情形,利用光學顯微鏡、電子顯微鏡及聚合�○s鎖反應配合限制�﹞軉q長度多型性研究海獸胃線蟲生物學,同時探討海獸胃線蟲經加馬射線後的致死率及生物學,期望對海水魚中最普遍的人畜共通寄生蟲-海獸胃線蟲有進一步的瞭解。研究發現白腹鯖、真�邥M白帶魚中海獸胃線蟲的平均感染率依次在為91%(66/72)、39%(28/72)和89%(64/72);平均密度依次為39條、2條和62條;平均豐度依次為62條、3條和80條。以形態學特徵和比對rDNA序列鑑定台灣基隆嶼海域的海獸胃線蟲為同一屬的寄生線蟲。魚體內海獸胃線蟲經加馬射線照射劑量分別為0 kGy (對照組)、5 kGy和10 kGy,照射後的海獸胃線蟲致死率評估為80%和83%,掃描式電子顯微鏡觀察微細構造變化,10 kGy加馬射線照射導致蟲體表面的損傷。研究結果顯示聚合�○s鎖反應配合限制�﹞軉q長度多型性,可以作為快速鑑定寄生蟲種類的工具,加馬射線照射海獸胃線蟲的抑制活性劑量為10 kGy。
Anisakis simplex is a parasite in marine fishes and mammalian, and may infect human being to induce Anisakiasis through seafood . The purpose of the work was to investigate the prevalence, mean intensity and abundance of the parasites (Anisakis spp.) in Trichiurus lepturus, Scomber japonicus and Trachurus japonica in the ocean around Keelung isle, Taiwan. By using light microscopy, scanning electron microscopy and molecular biotechnology, the biology of A. simplex and the effect of gamma-irradiation on the parasite control were studied. The prevalence of the parasite in T. lepturus, S. japonicus and T. japonica was 91%(66/72), 39%(28/72) and 89%(64/72), respectively. The mean intensity of the parasite in T. lepturus, S. japonicus and T. japonica was 39, 2 and 62, respectively. The abundance of the parasite in T. lepturus, S. japonicus and T. japonica was 62, 3 and 80, respectively. With PCR - based restriction fragment length polymorphism method and phylogenetic analysis, Anisakis spp. in the ocean around Keelung isle were separated two groups. Gamma-irradiation (5 and 10 kGy) offered a good control on the parasites. A. simplex with 10 kGy gamma-irradiation showed skin injuries. However, the nutrition of fishes irradiated was not significantly changed. Irradiation is a good option for controlling Anisakis spp. although the biology of the parasites needs further studies.
