情緒上的壓力、過度飲酒、過度飲用含咖啡因的飲料、幽門螺旋桿菌的感染及服用非類固醇抗發炎藥物等原因都會引發消化性胃潰瘍。許多研究指出西洋參具有抗氧化活性、降低肌肉細胞的損傷、調節免疫活性、降低低密度脂蛋白過氧化及降低高血糖的活性等功能,但對於胃潰瘍方面的研究具有較少的研究。本研究探討西洋參對酒精誘發的胃粘膜損傷時的保護作用。將32隻雄性Wistar 大白鼠,隨機分作4組:西洋參低劑量 (250 mg/kg)、中劑量 (500 mg/kg)、高劑量 (1250 mg/kg) 以及控制組 (10 mL/kg),各組8 隻。隨後60 分鐘再以70%乙醇 (10 mL/kg) 誘導大鼠的胃損害,4小時後將大鼠犧牲。結果顯示西洋參能夠有效抑制酒精所誘發的IL-1β 及COX-2 而達到抗發炎效果。除此之外,西洋參的補充能夠顯著增加Bcl2、Bcl-XL及pBad 的表現,抑制t-Bid、Cytochrome c、caspase 9、caspase 3等凋亡蛋白的表現量。因此,本研究結果發現,西洋參的補充能夠有效抑制發炎反應及細胞凋亡路徑中的粒線體路徑機制,來達到對胃黏膜具有保護的功能。
Emotional stress, heavy drinking, caffeinated drinks, infection of Helicobacter pylori and ingestion of non-steroidal anti-inflammatory drugs can caused gastric ulcers. Several studies reported that American Ginseng have various pharmacological activities including antioxidant activity, reduction of skeletal muscle cell membrane damage, immunomodulating activity, reduction of low density lipoprotein oxidation and reduction of hyperglycemic activity. But in the case of gastric ulcer, just has a few of study about it. In this study, we investigated the protective mechanism of American ginseng in ethanol-induced gastric apoptosis. 32 male Wistar rats, were randomly divided into four groups : Control (tap water 10 mL/kg) , American ginseng low dose (250 mg/kg), medium dose (500 mg/kg) and high dose (1250 mg/kg) , each group of 8 rats. After 60 min, 70 % ethanol was given orally to each animal at a dose of 10 mL/kg to induce gastric ulceration. Four hours after ethanol administration, the animals were sacrificed. Our results showed that American ginseng can significantly inhibited IL-1β and COX-2 to achieve the anti-inflammatory effect. In addition, American ginseng also can significantly increased in Bcl2, Bcl-XL, pBad activities and inhibited in t-Bid, Cytochrome c, caspase 9, caspase 3 activities. In conclusion, our results found that American ginseng can significantly inhibited inflammation and apoptosis of mitochondria pathway.
