由於魚類生活於水中,因而以血液檢測來評估水生生態環境中潛在的基因毒性時,被認為是很好的模式。唯許多有關水生環境汙染之研究多著重於淡水魚或硬骨魚類,軟骨魚受污染之研究極為罕見。為了解台灣船舶及漁業常用之抗附著劑之三酚基錫(triphenyltin, TPT)對軟骨魚類血液毒性現象之影響,本研究以斑竹狗鮫(Chiloscyllium plagiosum)為指標魚種,探討 TPT對其血液之細胞及基因毒性,紅、白血球數量的影響,並利用血液微核發生率,作為早期發生污染的無傷害檢測。

斑竹狗鮫經 TPT處理以後48小時半致死濃度為157.80 μg/l。隨TPT濃度增加,紅血球數的百分比有先增後減的趨勢,經檢定後發現TPT的加入會導致斑竹狗鮫之紅血球數量降低。白血球隨濃度的增加其數量百分比有顯著增加的趨勢。嗜伊紅性白血球數量的觀察隨濃度增加其數量也有增加的趨勢。

經 TPT處理後,紅血球出現染色體分布不平均,細胞質出現空泡以及細胞核消失等細胞型態之變化。空泡化及細胞核的變形與對照組呈顯著差異(p<0.05)。經 SEM觀察發現經 TPT處理後的血球有膨大、細胞變形成紡錘狀的情況。經統計檢定後,各處理組的微核發生率均與對照組呈顯著的差異,且在高濃度處理組出現雙核的頻率也增加。細胞核之比例不會因處理組濃度的增加而與對照組有差異。由本實驗可知,TPT亦會導致海洋軟骨魚類細胞及基因毒性之影響。
Fish can be useful for evaluating the cytotoxic and genotoxic potential of aquatic ecosystem as the directly contact with water. Most of these studies focused on freshwater and/or teleost species. Only a few reports have been concerned with Chondricythyes. In order to understand the toxic effects of a popular antifoulig paints (triphenyltin, TPT) to whitespotted bamboo shark (Chiloscyllium plagiosum), a coastal shark which is also very popular as human seafood, were studied in the present study. The changes of red blood cell counts, white blood cell counts, the morphological change of erythrocytes and micronucleus test as indicator for non-destructive tests.

The LC50 value of TPT to Chiloscyllium plagiosum is 157.80 μg/l. Exposure to sublethal levels of TPT resulted in significant decrease for red blood cell count compared with the control group. The counts of white blood cell were significant increased.

The morphology change of the erythrocytes (eg. altered cell, anisochromasia, vacuolation and enucleated cells) after TPT exposured for 48hr. The vacuole of the altered erythrocyte are the most common observed altered cell. Micronucleus frequency profiles showing dose-response relationship after TPT exposure for 48hr. There was significant increasing between control group and treatment groups. The erythrocyte nucleus ratio of cytoplasm between the control group and treatment groups was showing no significant difference. The result shows that TPT can induce erythrocyte and nucleus extraordinary to marine Chondricythyes.
