中文摘要
本研究主要探討卻老去皺美容複方中,單一或全部藥材以水或25﹪乙醇萃取的萃取物對抗氧化力與對皮膚纖維母細胞增生與光損傷之保護能力。抗氧化功能測定項目包括清除DPPH自由基、清除超氧陰離子、清除過氧化氫及螯合亞鐵離子效力,複方中所包含的中草藥種類:零陵香、白附子、香附子、川芎、甘松、白芷、青木香、茯苓與羊髓等藥材,其結果如下:
卻老去皺之單方或複方藥材以水及25﹪乙醇兩種不同溶劑萃取後,以羊骨髓與川芎萃取率較佳,茯苓、白蠟萃取率較差。由實驗結果得知,(1)抗氧化能力皆隨藥材萃取物隨濃度增加而提升,又以25%乙醇萃取物比水萃取物有顯著之抗氧化力;以植物性萃取物比動物性萃取物之抗氧化能力顯著較佳。
(2)中草藥水萃取物對於皮膚纖維母細胞之生長的影響,發現於 0.039 ~ 5 mg/mL時,以白芷、青木香與複方之萃取物顯著具有促進細胞增生之功效;25%乙醇萃取物於濃度0.039 ~ 0.625 mg/mL時,則以零陵香、川芎與甘松之萃取物,具有顯著促進細胞增生之能力。(3)皮膚纖維母細胞經紫外線B照射後,以添加水或25%乙醇之複方或單方之萃取物,於濃度0.031 ~ 0.04 mg/mL是保護皮膚纖維母細胞抵抗紫外線之最適濃度。
目前的實驗結果顯示,卻老去皺之單方萃取物之抗氧化、促進皮膚纖維母細胞增生與增強其防護紫外線損害之功效,皆比複方萃取物具有較佳效果,至於各個單一藥材之間是為相乘或抑制作用,仍須進一步探討,上述萃取物對皮膚系統之效果,因實驗設計的關係無法評估。
Abstract
The aims of the study were to characterize the capacity of antioxidation and enhancing effect on the proliferation and the protection of fibroblast from by photodamageing with the extracts of individual remedy or chyueh laau chyuh jouh miahn gau (Anti-skin aging, Anti-wrinkle cream) of water or 25% ethanol. Antioxidant assays include reducing power assay、1, 1-diphenyl-2-picrylhydrazyl (DPPH) scavenging、Superoxides amino (O2-‧)、hydrogen peroxide (H2O2) and chelating ferrous ions capacities . The complex herbs remedies included Ociumum basilicum L., Aconiti coreani tuber, Cyperi rhizome, Ligusticum wallichii franch, Vardostachys chinensis batal, Angelica dahurica benth, Aristolochia mollissima hance, Poria cocos (schw.) wolf and Sheep’s bone marrow. The results obtained are as following:
Extraction efficiency of Ligusticum wallichii franch and Sheep’s bone marrow are high on than other remedies howerer extraction efficency of Poria cocos (schw.) wolf. and Angelica dahurica are lower than others.(1)The antioxidation capacity of extracts increased with their concentration. The antioxidation capacity of extracts with 25% ethanol was better than that of with water so as the extracts of plants were batter than from animals.(2)Water extracts of Vardostachys chinensis batal, Angelica dahurica benth and Fu fang at the concentration between 0.039 ~ 5 mg/mL promoted the cell proliferation significant. The concentration of 25% ethanol extracts of Ociumum basilicum L., Ligusticum wallichii franch, Vardostachys chinensis batal, Angelica dahurica benth between 0.039 ~ 0.625 mg/mL promoted the cell proliferation significant.(3)The optimum concentration of the extract that shower protect on photo-damaging by UVB was 0.031 ~ 0.04 mg/mL.
The extract of individual remedies were better than Fu Fang in the capability of antioxidation, proliferation of human skin fibroblast and protection of photo damaging by UVB. Are there any synergistic action or inhibition among the extracts of individual remedies and/or Fu Fang needed further studies. The effect of those extracts on human skin system could not conclude due to the design of experiment.
