苦楝(Melia azedarach Linn.)屬於楝科(Meliaceae)為本土速生樹種,幾乎廣泛分布全臺,生育地條件複雜,經過長時間自然選擇,形成許多具有適應不同氣候的地理生態型。本研究即應用高效能液相層析法(High Performance Liquid Chromatography,HPLC)與簡單序列重複(Inter-Simple Sequence Repeat,ISSR)分子指紋技術探討其化學成分含量與遺傳多樣性。在臺灣地區不同氣候區由北至南所設之18個樣區,130個樣木中,HPLC成份分析結果顯示,各氣候區間苦楝族群化學成分之川楝素(toosendanin)含量有明顯的差異。使用8組ISSR引子與樣木進行PCR擴增。其中多型性條帶有44條(66.67%)。族群遺傳變異分析(POPGENE)結果計算族群Nei’s基因歧異度值(H)為0.3930,各地區族群介於0.1285-0.3559。族群間之遺傳分化係數 ( Gst )為0.4153,族群基因流( Nm )為0.7040。分子變方分析(AMOVA)結果,顯示氣候區間之變方成分為19.30%(p<0.001),氣候區內族群間之變方成分是14.03%(p<0.001),氣候區內的族群內個體間變方成分是66.67%(p<0.001),說明苦楝個體間的遺傳變異是苦楝族群總變異的主要來源,但氣候影響就已佔其近20%,不同氣候與族群間已有分化趨勢。探討各生育地族群間的遺傳分化及基因交流情形與不同氣候區之關係,由mantel結果其遺傳變異與平均氣溫、平均年日照與地理距離有顯著差異。综言之,各氣候區之苦楝族群基因流動並不暢通,已有分化現象,可能與花部構造導致繁殖系統多為自交、種子有毒,影響動物傳播及長期在不同地理環境下的天擇有關。
Melia azedarach Linn. belonging to Meliaceae, which was fast-growing native tree, widely distributed in almost all of Taiwan, the complexity of habitat conditions, after a long period of natural selection, the formation of many different geoecotype. The study was applied HPLC and ISSR molecular fingerprint technology to explore the chemical composition and genetic diversity of content. In different climatic regions in Taiwan from north to south by the set of 18 sample areas, 130 samples of the M. azedarach , HPLC analysis of chemical compositions results showed that was a significant variation in toosendanin content among the regions. A total eight primers were used and 44 polymorphic sites were obtained. Population genetic ( POPGENE ) analysis revealed that total gene diversity ( H ) was 0.3930 and genetic index ( Gst ) was 0.4153, while gene flow index (Nm) was 0.7040. Analysis of molecular variance ( AMOVA ) revealed that the variance component between population regions was 19.30 % (p< 0.001 ), among populations within regions was 14.03 % (p<0.001 ), and among individuals within population was 66.67 % (p<0.001 ). The result indicated that the major variation existed among individuals within populations; however, there was a significant genetic differentiation existed among regions and among population within regions, the climate has been accounted for nearly 20%. Explore the habitat of the genetic differentiation among populations and gene flow situation between different climate regions. From the mantel test know the results were significant differences in genetic variation and average temperature, average annual sunshine and geographical distance. In conclusion, the gene flow among populations was relatively restricted. Floral structure, poisonous seed and geographic climate segmentation may be a significant factor to the genetic differentiation among these populations.
