Carbon isotope ratio analyses for propane and butane have been reported for years and become part of routine tools for geo -chemical studies. Recently, Chung et al. proposed an ideal equation to explain the kinetic isotope effects of petroleum- forming process.However so much assumption had made before deriving this ideal equation. The aims of this study is to search the directly evidence to support this fact . Thus , propane and butane were oxidized by methane monooxygenase catalyzed reaction to the corresponding 2- propanol and 2-butanol . The 2-propanol will be conducted to the iodoform reaction to yield ethanol . The intramolecular C-13 isotope ratio can be calculated from assay of ethanol and 2- propanol. Similarly,the butane molecule were analyzed. Because short of methane monooxygenase , meanwhile the most works were focused on these 2-propanols and acetone obtained from industrial company.The result indicated that the 2-propanol and acetone from different sources might have different C-13 stable isotope ratio. For those with similar bulk data molecules can be further identified by comparing their intramolecular isotope ratio of corresponding carbons.
