巴拉金糖(palatinose)又稱為為異麥芽酮糖(isomaltulose),為存在於蜂
蜜或蔗汁中之雙糖(.alpha.-D-glucopyranosyl-1,6-fructofuranose),其
甜度雖僅約為蔗糖之42%,但由於具有抑制由蔗糖生合成水不溶性葡聚糖
(gl-ucan)之作用,而可防止齲齒之形成,因此近年來此種生物技術產品
已普遍受到食品加工界之重視。本研究針對巴拉金糖之工業生產進行探討
,獲致如下成果:Klebsilla planticola NTU-356可由蔗糖以高活性生產
巴拉金糖。使用每公升含70公克蔗糖、50 毫升大豆粕水解液、5 公克氯
化鈉、2.5公克磷酸氫二鉀之培養基,以起始pH 7.0,攪拌速度200 rpm,
通氣量1 vvm之培養條件在小型醱酵槽中之巴拉金糖生成活性為16.5 U/mL
,而進一步放大規模至1,000公升中間工廠醱酵槽培養時,其巴拉金糖生
成活性為17.6 U/mL。以 3%海藻酸鈣固定化菌體,在生物反應器中連續
生產巴拉金糖,使用50%蔗糖為基質,反應溫度40℃,基質流速為0.1
sv/h時,巴拉金糖之產率可達81.3%,並可連續生產12日。視用途而定,
可使用有機溶劑沉澱法或全量固形化法將液態巴拉金糖精製成固態粉末以
便儲存與運輸;有機溶劑沉澱法所得之結晶純度高,而全量固形化法之回
收率幾乎達 100%,純度則與原液之巴拉金糖含量相同。
Palatinose(.alpha.-D-glucopyranosyl-1,6-fructofuranose),some-
times referred as isomaltulose, exists in honey or cane juice.
The sweetness of palationseis about 42% of that in sucrose,and
yet it inhibits the formation of water insoluble glucan from
sucrose, therefore it can prevent cavity. In recent years, the
commercial production of palationse by transglucosylation has
been well re- cognized in the food industry. The objective of
this study is to provide a systematic study on the enzymatic
production of this noncariogenic sweetner, and develop an
economical bioprocess to produce palatinose. In this study,
following subsjects were com- pleted: Klebsiella planticola
NTU-356 is a high palatinose-produ- cer from sucrose.The enzyme
production was optimized by submerged cultivation in a jar
fermenter with initial pH at 7.0, aeration at 1 vvm, agitation
at 200 rpm. Under these conditions the pala- tinose-producing
activity was 16.5 U/mL. Furthermore,the activity was 17.6 U/mL
in a 1000-liter pilot-scale fermenter in a medium containing
70 g/L sucrose , 50 mL/L soybean meal hydrolysate , 5 g/L
sodium chloride and 2.5 g/L dipotassium phosphate. Produc-
tion of palatinose was also performed continuously with
immobi- lized cells. The cells were entrapped in 3%calcium
alginate gels and were packed into a pilot-scale bioreacter.
The optimal condi- tions for operation were asfollows:
substrate concentration, 58% sucrose; space velocity, 0.1 sv/h;
operation temperature, 40℃ . The yield of palatinosewas 81.3%.
The bioreactor can be operated for 12 days without renewing the
gels. Palatinose was recovered by organic solvents with high
recovery and by total solid form with high efficiency. The
recovery of palatinose by total solid form was as high as 100%
and the composition of products was the same as that from
bioreactor.
