In order to develope a non-destructive technique to detect denitrification in soils, the piezoelectric quartz gas biosensor is taken a selected method here to detect gaseous intermediate metabolites of denitri- fication. The purpose of this studies was to establish a method that could analyze rapidly. The 9 MHZ quartz crystals were used in this study , and sealed injection method was applied to analyze gaseous intermediate metabolites of denitrification. The application of various chemically and biologically coated crystals were tested, and interference caused by the humidity of gas, so the relative humidity of sealed bottle was controlled at 20% during the mea- surement. The results indicated that the crystals coated with Thiosphaera pantotropha membrane showed better response than the others examined. And the data could fit the Langmuir equation. A linear cali- bration over the range of 0.1∼20 ppm of nitrous oxide was obtained and the detection limit of the technique was determined to be 0.1 ppm . The amount of coating with T. pantotropha membrane would effect the detecting range, response frequency and response time. The coating with 107.14.sim.42.86 .mu.g on crystal was very effective for practical applications.
