杏仁核(amygdala)與情緒記憶有密切關係。然而杏仁核是否直接參與記憶的形成並且永久貯存情緒記憶,目前仍無定論。大腦皮質的島狀皮質(insular cortex)、內側前額皮質(medialprefrontal cortex)及嗅緣皮質(perirhinal cortex)接受大量來自杏仁核的神經投射,有可能涉及情緒記憶的長期貯存。本論文之目的便在於探討杏仁核及大腦皮質在情緒記憶的形成及長期儲存中所扮演的角色。
本研究採用抑制性逃避學習作業(inhibitory avoidance task)來測量情緒記憶。大白鼠事先接受杏仁核或島狀皮質、內側前額皮質及嗅緣皮質之注射導管埋值。實驗一中,大白鼠在訓練前進行紋終(stria terminalis,ST) 毀除或假性毀除手術,訓練後立即在NMDA或APV處理下,學習後一天的記憶表現會產生明顯的缺陷。此記憶損害的作用在ST毀除後依然存在。實驗二,大白鼠於測試之前五分鐘以2%局部麻醉劑lidocaine壓抑島狀皮質、內側前額皮質或嗅緣皮質的神經活動,對於訓練後一天的記憶表現均不造成明顯的影響;但以lidocaine分別或同時壓抑島狀皮質與內側前額皮質,2l天久遠記億的表現均受到明顯的損害。而壓抑嗅緣皮質對於21天久遠記憶的損害作用只有在大白鼠被重複測試之後方可表現。實驗三在訓練之前以鋼絲刀將杏仁核與其附近大腦皮質之聯絡神經在外囊(external capsule)處切斷,發現並不影響訓練後21天久遠記憶的表現。
綜合以上之結果,可知杏仁核的確參與了情緒記憶的形成,至少其內之麩胺酸傳導素系統(glutamatergic system)包含在情緒記憶形成時所涉及的神經迴路中。情緒記億的神經痕跡形成之後,可能移轉至島狀皮質及內側前額皮質進行長期貯存。然而,外囊中杏仁核與皮質之聯絡神經可能並非情緒記億移轉至長期儲存之處所必經之路徑。
The present study investigated the roles of the amygdala and its connected cortical areas in formation and storage of affective memory. Male Wistar rats were implanted with cannulae bilaterally into theamygdala, insular cortex, medial prefrontal cortex or perirhinal cortex.They were trained on the one-trial inhibitory avoidance task and tested for memory 1 or 21 days later. In Experimental I, amygdala-implanted rats received radiofrequency lesions of the stria terminalis (ST) or sham peration. Immediately after training, 1.0 μg of NMDA or 1.25 μg of PV was infused into the amygdala. The 1-day retention test indicated that both NMDA and APV impaired memory and this amnesic effect was impervious to ST lesion. In Experiment II & III, cortical-implanted rats were tested 1 or 21 days after training. Pretest infusion of lidocaine into the various cortical regions had no effect on retrieval of the 1-day memory. However, pretesting infusion of lidocaine into the insular or medial prefrontal cortex, either alone or in combination, impaired retrieval of the 21-day memory. Pretest infusion of lidocaine into the perirhinal cortex had o effect on the 21-day memory retrieval unless the rat had been tested previously. In Experiment IV, the amygdala-cortical connection in the external capsule was transected by a wire knife prior to the inhibitory avoidance training. The results indicated that the transection had no effect on memory performance in either 1 or 21-day test.
These findings, taken together, suggested that the neural trace of affective memory may be formed in a neural circuit including the amygdala. However, the memory trace may translocate to the insular and medial prefrontal cortex sometime after its formation for long-term storage. Fibers in the external capsule do not seem to be involved in the translocation of the inhibitory avoidance engram.
