在癌症治療方面,放射治療和藥物治療是二種主要之治療方式,臨床上為
治癒癌症,常必須提高藥物或游離輻射的劑量,如此常會對 體的正常組
織或器官造成極大的傷害。若將化學藥物和游離輻射合併使 處理癌症,
往往會有加成作用(addition)或協同作用(synergism),而j大降低單獨處
理時化學藥物或放射治療所使用之劑量,如此不但可減少鴷膨`組織的傷
害,同時又可 治癒癌症。本研究的主要目的在探討,以d-limonene (帖
烯;0 mM, 0.5 mM,mM, 1.5 mM, 2 mM和3 mM)或游離輻射線(銫-137;0
Gy, 1Gy, 2 Gy, 4 Gy, 8 Gy和12 Gy) 處理人類子宮頸癌細胞(HeLa
cells),就兩者個別B理或合併使用時對HeLa細胞之殺傷作用,分別從三
方面進行探討:(1)以H不同濃度d-limonene處理HeLa細胞1~3小時後,其
內還原態和氧化態麩胱? (reduced glutathione,GSH;oxidized
glutathione,GSSG)含量之變①C(2)以1 mM和1.5 mM d-limonene處理HeLa
細胞後,對細胞週期之影響C(3)以不同濃度的d-limonene和不同劑量的游
離輻射線單獨或合併處理eLa 細胞後,細胞存活的情形。合併處理的方法
有三種情況:(a)先以.75 mMd-limonene作用細胞三小時後,立即施予輻
射線照射;(b) 先以輻g線照射 mM d-之後,立即施予0.75 mM d-
limonene 處理三小時;(c)在.75limonene處理三小時的過程中,同時施
予輻射照射。 結果顯示:(1) GSH會隨著d-limonene作用時間的增加和濃
度的提高而下降。1 mM-limonene 處理3小時後,降低GSH含量至控制組
之77 %;2 mM d-limoneneB理3小時後,則降低至54 %。而GSSG之含量,
卻會隨著d-limonene作用時〞獐W加和濃度的提高而上升。(2) d-
limonene會阻斷HeLa細胞之細胞週薊熄i行,使其停留在G1期,而無法進
入S期,因而抑制DNA之合成;細胞L法合成DNA將影響其生長,甚至造成其
死亡。(3) 合併d-limonene和游離蝞g線對HeLa細胞作用後發現,以0.75
mM d-limonene配合1 Gy, 2 Gy或Gy的輻射增敏作用最大,對HeLa細胞具
有協同殺傷作用,可達到單獨使? mM d-limonene或12Gy游離輻射線照射
的效果。而其三種處理之方式,蚳銗H先用d-limonene作用細胞三小時後
再施予輻射線照射之效果最佳。?.75 mM d-limonene與4 Gy游離輻射線
配合使用時,其對細胞之殺傷力P其它二種方法有顯著差異(p<0.05)。

Combination of radiotherapy and chemotherapy usually has better
tumor killing and less normal tissue damage effects due to
addition or synergism of the two treatments. In this we
investigate the killing effect of d-limonene alone (0 mM, 0.5
mM, 1 mM, 1.5 mM, 2 mM and 3 mM) or combined with ionizing
radiation (IR; Cs- 137 irradiator, dose rate 7 Gy/min;
irradiated doses are 0 Gy, 1 Gy, 2 Gy, 4 Gy, 8 Gy, and 12 Gy)
in HeLa cells. The following three subjects were studied.
Firstly, after 1 to 3 hours treatment of d-limonene, the change
of intracellular contents of reduced glutathione (GSH) and
oxidized glutathione (GSSG). Secondly, the cell-cycle effect of
d-limonene. Thirdly, the survival of HeLa cells after d-
limonene alone or combined with IR. There are three conditions
for combination treatments: (i) cells were treated with 0.75 mM
d-limonene for 3 hours, then irradiated; (ii) cells were
irradiated first followed immediately with 0.75 mM d-limonene
for 3 hours; and (iii) cells were treated with 0.75 mM d-
limonene and simultaneously irradiated . The results showed
that the GSH level decreases linearly with the increase of the
treatment time and concentration of d-limonene. After 3 hours
treatment of d-limonene, the GSH contents were reduced to 77 %
and 54 % of that of control for 1 mM and 2 mM, respectively. On
the contrary, GSSG level increases with the increase of the
treatment time and concentration of d-limonene. Cells treated
with d-limonene are arrested at G1. Besides, the cell growth
could be completely suppressed by 2 mM d- limonene. For
combination treatments, there is more better killing effect
among the three protocols when cells were treated first by d-
limonene, and we found the radiosensitization of d-limonene is
synergistic when combined with 1 Gy, 2 Gy or 4 Gy. radiotherapy.
