在本研究中探討Acinetonacter radioresistens所生產的嗜鹼性脂肪酉每之部分特性及其醱酵條件。A. radioresistens所生產的脂肪酉每在37℃及pH10時有最大反應活性。金屬離子中之Cu2+、Co2+、Zn2+會抑制酵素活性,結果也顯示在SDS及Triton X-100等界面活性劑存在下易使酵素失活。A. radioresistens生產脂肪酉每的最適溫度及pH值為30℃及pH7,同時攪拌速率增加可提高菌體生長速率及脂肪酸生產,在含有 1%tryptone、0.5% yeast extract、1%氯化鈉、0.1%橄欖油、0.1%氯化銨和1%正十六烷的培養基培養時,有最高的脂肪酉每活性(17U/mL)。最大脂肪酉每活性在對數生長末期出現,此時伴隨產生大量泡沫及蛋白酸的生成。在連續醱酵之不同稀釋率中,當稀釋率為0.1h-1時,有最高的脂肪酉每活性。而且此時菌體不再分泌蛋白酉每,所得比酵素活性為批次的兩倍,因此連續醱酵有助於A. radioresistens生產耐鹼性脂肪酉每。
The production and partial characterizations of an alkalophilic lipase from Acinrtobacter radioresistens were investigated in this study. The lipase was most active at 37℃ and pH10.5. Cu2+,Co2+, and Zn2+ were found to inhibit the lipase activity. The result also showed that SDS and Triton-X decreased the stability of lipase. It was indicated that the optimum temperature for lipase production was 30℃. The highest activity of lipase was obtained when cells were cultivated at pH7. The result also showed that both cell growth rate and lipase production increased as increasing in agitation speed. The optimal medium for lipase production consisted of (w/v%): tryptone, 1 ; yeast extract, 0.5; NaCl, 1; NH4Cl, O.1; oliver oil, O.1; and n-hexadecane, 1, which resulted in 17 U/mL of lipase activity. In addition, the highest activity of lipase was found at the late exponential growth phase, where large amount of bubble formed and the protease was began to secrete into the broth. In the continuous operation, dilution rate of O.1h-1 resulted in the highest activity of lipase among the other dilution rates. The specific lipase activity obtained from continuous culture was duple higher than that from batch operation. Additionally, protease was not found in the continuous culture. Therefore, continuous operation was more favorable for the production of A. radiorestens lipase.
