Gardenia jasminoid Ellis is a commonly used Chinese herbal drug which is used in traditional Chinese medicine as an antiphlogistic, diuretic, laxative, choleretic, and hemostatic in the treatment of traumatosis by external application. In this research, a high-performance liquid chromatographic method for the determination of six components was developed within 60 min. This method was carried out by using a gradient solvent system of phosphate buffer-acetonitrile-methanol and detecting at 240 nm and 440 nm. The effects of column selectivity, phosphate concentration and organic modifier concentration of the mobile phase on the retention and separation of the solutes were also studied. Huang-lien-chieh-tu-tang, a Chinese herbal preparation, is known to consist of four herb drugs. In this part, a total of 14 components which included 6 quartemary alkaloids in Coptidis rhizoma and Phellodendri cortex and 4 flavones in Scutellariae radix and 4 glucosides in Gardeniae fructus could be determinated by a LC method using gradient solvent system that contained phosphate buffer. The effects of mobile phase and column selectivity were also discussed.
