A high-performance liquid chromatographic method was developed to separate ten bioactive constituents in Artemisiae Capillaris Herba. The method was carried out by using a gradient solvent system of phosphate buffer-methanol-acetonitrile, Cosmosil 5Cis-MS column and detecting at 280 nm, and the contents in Artemisiae Capillaris Herba extract could easily be determined within 85 minutes. The relationship between the external appearance and the quality of the herb drug had been investigated, and the differences among the samples of Artemisiae Capillaris Herba and Origani Herba were also evaluated. The effect of column, mobile phase and precolumn on the separability was studied. Yin-chen-hao-tang, a Chinese herbal preparation, is known to consist of Artemisiae Capillaris Herba, Gardeniae Fructus and Rhei Rhizoma. A high-performance liquid chromatographic method for sitmultaneous determination of five components in Artemisiae Capillaris Herba, three in Gardeniae Fructus and five in Rhei Rhizoma was developed. Detecting at 250 nm with a Cosmosil 5C18-MS column and a linear gradient elution system consisted of KH2PO4 methanol and acetonitrile was found to be able to determine thirteen constituents within 80 minutes. Using this method, various samples of Yin-chen-hao-tang were evaluated and compared. The effect of column, mobile phase and precolumn on separation was also studied.
