Gelatinase是MMP家族的子群,含有兩種蛋白:gelatianse A( 72-kDa
type IV collagenase, MMP-2)和gelatinase B( 92-kDa type IV
collagenase, MMP-9).。它們是可以分解多種細胞外基質的分泌性蛋白,
可以和TIMP形成非共價性的結合並被TIMP抑制其活性。在生殖系統中,
gelatinase被認為和排卵、著床、懷孕等需要細胞外基質分解使組織得以
重組來因應生理變化的現象有關。在人類的子宮內膜中發現gelatinase A
和gelatinase B mRNA的表現和蛋白活性都受到黃體素的抑制。本論文嘗
試藉觀察在動情週期各階段時gelatinase的mRNA和蛋白在小白鼠子宮的表
現情形來瞭解在小白鼠子宮中gelatinase是否會受到荷爾蒙的調控。在
gelatinase B mRNA的表現上,我們利用北方墨點法和逆轉錄-聚合?@放大
反應(RT-PCR),結果顯示其mRNA表現很少且在動情週期可能無顯著的改
變。在蛋白的表現上,利用zymography觀察到有三種主要gelatinase出現
在老鼠子宮組織。而且105kDa gelatinase也有存在於子宮分泌液中。在
動情期、動情後期gelatinase的表現有增加的情形,進入動情休止期和動
情前期則有減少的趨勢。顯示gelatianse的活性在小白鼠子宮中可能受到
黃體素的抑制。因此gelatinase在小白鼠的子宮在動情週期各階段所產生
的組織形態變化上可能扮演某種角色。
Gelatiase is a subgroup of matrix metallproteinase. The members
of gelatinase are gelatinase A ( 72-kDa type IV collagenase,
MMP-2)and gelatinase B ( 92-kDa type IV collagenase, MMP-9).
They are secretory enzyme which can degrate many components of
extracellular matrix and they can form noncovalent complex with
tissue inhibitor of metalloproteinase that inhibit their
activity.Gelatianases were thought to play a role in
reproductive system such as ovulation, implantation, and
gestation, which often require extracellular matix remodeling.
And in human endometrium, the mRNA expression and activity of
gelatinase A and gelatianse B are inhibited by progesterone. In
this thesis, I tried to determined the gene expression and
protein activity of gelatinase A and gelatinase B in mouse
during estrus cycle to find that if they were regulated by
steroid hormone. The expression of gelatinase B mRNA were
investigated using North Bloting and RT-PCR. The gene expression
for gelatinase B in mouse uterus is very low and maybe is
constitutive during estrus cycle. In protein level, zymography
was used zymography and find that there are three major
gelatinases in mouse uterus. The activity of gelatinases is high
in estrus and metaestrus uterus. In diestrus and proestrus
uterus is low. And there is expression of 105kDa gelatinase in
uterus fluid. This finding indicated that the activity of
gelatinase is inhibited by progesterone in mouse uterus, and
implied their expression have specific roles in the tissue
growth and remodeling processes occuring in mouse uterus during
estrus cycle.
