The possibility of using oligonucleotides to regulate gene
expression at the DNA/RNA level has recently led to renewed
interest. However, protonation at N-3 of cytosine is required in
order to form two H-bonds with guanine of the DNA strand. This
limits the potential use of such oligomers as anticode
inhibitors under physiological conditions. According to the
molecular modeling, 8-oxoadenine can interact in a speciific
manner with GC base pair without protonation; hence this
modified base may be a useful alternative to cytosine for use in
DNA triplex formation at neutral pH. In addition, it has been
reported that certain alkoxy modifications in the position 2'-
of the ribose ring significantly increase the binding aaaffinity
of the oligomer to its target sequence, and provide the
reesistance to ribonucleases. In this thesis, it has been
discussed that the synthetic strategies and the reaction
conditions of deerivatives of 8-methoxy(-2'-deoxy-) adenosine,
which are precursors of the corresponding 8-oxo(-2'-deoxy-)
adenosine.
