傳統的魚肉中偵測多氯聯苯(PCBs)含量的方法,主要是以皂化法破壞魚類組織,再以液─液萃取方式將多氯聯苯萃取出,其過程不僅耗時且需要大量溶劑。本研究主要是以微波萃取法,萃取魚肉中之多氯聯苯含量,並嘗試以不同之淨化(矽膠 、矽酸鎂及礬土等)方式,利用GC/ECD測定,以回收率作指標,比較微波萃取與魚肉皂化之效果。微波法回收率80%以上大於皂化法回收率75.6%。
微波條件80%W→ ramp 5min→115℃(hold 10min),以魚肉參考樣品CARP-1之PCBs平均回收率為93.2%,''因此證明此微波條件微波效果相當好 。
本研究主要偵測魚類中常含之多氯聯苯為(IUPAC No.18, 31, 44, 52, 101, 110, 138, 153, 170, 180)等10種,測出10種PCBs之方法偵測極限範圍為2.2∼12.3pg,最後進行高污染性魚類之分析,其中二仁溪污染流域魚類,以豆子魚、大眼海鏈、吳郭魚較高,泰國鯉魚較低,10種PCBs含量約在0∼172.5ng/g,而PCBs種類以PCB-1O1、PCB-11O、PCB-153及PCB-180含量較高。而淡水河之污染魚類以大眼海鏈、吳郭魚含量較高,豆子魚較低,10種PCBs含量為0∼16.6ng/g,其含量遠較二仁溪為低,二仁溪河口之花身仔魚及豆子魚與基隆河八堵區段及成功橋之吳郭魚,測得少量之PCBs含量約為0.10∼2.88ng/g,PCBs有PCB-44、PCB-11O、PCB-138、PCB-153、PCB-170、PCB-180及PCB-182/187等。
The traditional methods for detecting the amount of polychlorinated biphenyls (PCBs) in fish are mainly to destroy the fish tissue by sponification and then to extract the PCBs by liquid-liquid extraction. This process not only is time- consuming but also requires a large amount of solvent. Therefore, the goal of this study is primary to detect the amount of PCBs in fish using microwave extraction and different cleanup procedures, such as silica, Florisil and alumina, etc. The concentration of PCBs congeners can be determined by using gas chromatography/electron capture detector(GC/ECD) and indicating of quantitative recovery, and comparison of the results between microwave extraction and sponification. The results show that quantitative recovery from microwave extraction is above 80%, which is larger than that of sponification, (75. 6%. )for 10 PCBs congeners.
The procedures for microwave extraction was that temperature was increased to 115℃ in 5 minutes by 80% microwave power and then hold for 10 minutes. The average quantitative recovery of 9 PCBs congeners is 93.2% in certified reference material CARP-1 of fish. Hence, it is evident that the result of microwave extraction is quiet good.
This study is mainly to determine 10 PCBs congeners, i.e., IUPAC No.18, 31, 44, 52, 101, 110, 138, 153, 170, 180, spiked in fish and then to extract PCBs using microwave energy as well as the concentrations of PCBs is in the range of 2.2 to12.3 pg. Finally, we also found that the concentrations of PCBs of the fishes, such as Lizza carinata、Megalops cyprinoides and Tilapia in highly-polluted water (Er-Jen stream)have more than that of PCBs in fish as Channa sp. The amount of 10 PCBs congeners contained in fish from Er-Jen stream are ranging 0 to 172.5 ng/g. Among 10 PCBs congeners, the concentractions of PCB-101、PCB-110、 PCB-153、PCB-180 can be found more than that of PCBs. The concentractions of PCBs congeners of the fishes in Tamshui river are ranging 0 to 16.6 ng/g, Megalops cyprinoides and Tilapia contain more. The mount of PCBs congeners found in others areas can be detected less than 5.81 ng/g.
