An amylase which produeced maltotetraose and
maltopentaosefrom starch and glucose as the main products was
found inthe culture filtrate of a strain of TS-8. TS-8 strain
products the enzyme extracellulary when incubated in soybean
culture for 24 hours. Thebest composition of enzyme producing
media in shaking culture contained 2.5% starch, 1% soybean, 0.3
% yeast extract, 0.3% malt extract, and 0.03% MnCL2. The
maltopentaose amylase waspurified to homogeneity from the crude
extract of liquid culture by ammomium sulfate fractionation,
Fractogel DEAE 650 M, Fractogel phenyl 650M, and Fractogel TSK
HW-55 column. Finally, a 24 fold purification was obtained, and
the specific activity of the purified enzyme was 414 unit/mg
protein.The molecular weight of the enzyme determined by 12%
SDS-Polyacrylamide gel electrophoresis was about 66 kDa, and
from TSK HW 55 gel filtration estimated to be about 69 kDa, so
maltopentaose amylase is a monomer structure.The enzyme was with
an optimal reaction pH of 6.0 and was stable in the range of pH
6.0 to 8.0. The optimum reaction temperature of hte enzyme was
40 degree Cand stable in 50 to 60 degree C. Metal ions such as
Mg2+, Mn2+, Zn2+, Cu2+, Pb2+, Hg2+, and Fe2+ inhibited the
enzyme activity and the Ca2+, Co2+and Cd2+ could not inhibited
the enzyme activity. maltopentaose amylase could
hydroolyzed maltopentoase, maltohexaose, maltopentaose, starch
and produced glucose, maltose, maltotriose, maltotetraose,and
maltopentaose. According to the enzyme specificity of
hydrolyzing the maltooligosaccharides, and the enzyme was named
as maltopentaose amylase.
