The explants of the different habitual crops such as Lisianthus, Statice, and Gerbera were cultured on solid supporters, such as oasis, rockwool, multiblock rockwool, sponge, paper towel, or non-woven fabrics solid supporters to substitute the solidified media by agar, respectively, in order to improve the quality after long term transportation, to simplify the operation of transplanting, and to increase the survival rate ex vitro. The basal MS medium contained sugar 15 g/l, and IBA 2mg/l, NAA 0.5mg/l, or IBA 0.5mg/l when cultured prairie gentian , statice or gerbera, respectively. After in vitro culture for 5 weeks, when the supporters of sponge, multiblock rockwool , non-woven fabric or paper towel were used for prairie gentian, non-woven fabric and paper towel was used for statice, as well as paper towel was used for gerbera, the growth of explants on those solid supporters were better than those cultured on agar media. The volume of liquid medium was response to the kind of supporter used. When prairie gentian explants were planted on sponge, multiblock rockwool, non-woven fabric, or paper towel, the most suitable liquid level for culture was at 0cm (just saturation), 0.5cm, 1cm, or 1.5cm, respectively. When statice planted on paper towel, sponge, or non-woven fabric the best liquid level was at 0cm , 1.0cm, and 1.5cm, respectively. Sometimes there were a few of hydrosis of prairie gentian and statice explants, when cultured on liquid media. However, the container with permeable filters enhanced hardening and growth of prairie gentian and statice explants cultivated on sponge and paper towel.In addition, to establish the autophotosynthesis of explants, culture was conducted under low sugar medium in the medium and high photosynthetic photon flux density environment. Plantlets growth was the better when plantlets were cultured on the lower sugar concentration. It means when plantlets were grew on medium without sugar, the survival rate ex vitro was the highest. Moreover, liquid medium did not be sterilized if the solid supporters were clean.During light period, enriching 2 liter CO2 in growth chamber every 8 hours to increase CO2 concentration to 1700 ppm promoted the autophotosynthesis ability of plantlets. It means the plantlets of prairie gentian and statice had better quality in appearance and had lower density but larger ellipsoid and functional stomata.
