有機芳香族化合物經常被無意地排放至水域環境中,造成水域環境的衝擊 與毒害.為了做進一步的防治以及處理策略,必需先瞭解有機芳香族化合物 對水域環境的衝擊程度究竟有多少,意即我們必需先了解這些化合物的相 對毒性大小.我們首先以假單胞菌之生長受化合物影響知生長曲線實驗做 為參考,其次再利用兒茶酚雙加氧霉的受質消耗速率會受化合物影響的特 性,而以光譜觀察受質消耗初速率的改變,進一步利用動力學模式求取抑制 常數,以抑制常數做為比較相對毒性的依據.最後,更進一步將酵素實驗所 求得之結果,利用結構-活性定量關係探討化合物與酵素作用的機制. In order to understand the impact of these compounds to our environment,the relative toxicity should be est imate. Microbial methods have s hown to be potent processes to e stimate acute toxicity. Toxicity assessment of aromatic compound s to Pseudomonas putida isperfor med in the currend studies based on time-consuming microbial cul turetechniques and enzyme (metap yrocatechase) inhibition studies . The growth curvecan be plotted and it provides rough informati on of the results of the involve mentof aromatic compounds. In co ntrast,the enzyme method can giv e better results ofrelative toxi city (1/Ki) of aromatic compound s. Finally,log (1/Ki) was co rrelated with log P and 6,to sho w the quantitativestructure-acti vity relationship (QSAR) with in the compound classes,and the me chanism of the interaction betwe en aromatic compounds and enzyme was discussed.
