Lung cancers are increasing in Taiwan for the past 30 years. It is the leading cause of cancer death in women and the second in men, nonetheless the prognosis of lung cancer is generally poor. Furthermore, the epidemiologic characteristics of lung cancer in Taiwan are different from those in other parts of the world. Therefore, it is important to study lung cancer in Taiwan. LOH on chromosome 11 occurs frequently in lung cancer indicating that chromosome 11 may have a role in the pathogenesis of lung cancer. tsg101 was identified as a tumor susceptibility gene by homozygous functional inactivation of allelic loci in mouse 3T3 fibroblasts. The human TSG101 homologue was mapped at chromosome 11p15 and found to have intragenic deletion in breast cancer. To determine whether abnormal TSG101 expression is common in other tumors, TSG101 gene expression in 29 primary lung cancers and More than one abnormal transcript are found in the same samples. Sequence analysis demonstrates that there are at least fourteen distinct deletions. Six of them are existed in more than one sample. The most common deletion identified, from bp 154 to 1054, was identical to a deletion reported previously in breast cancer. The breakpoints of most transcript deletions coincide with genuine or cryptic splice site sequences, suggesting that the results from aberrant splicing. The results also demonstrate
