3,4,5-Trimethoxy benzoic acid(TMB) 類似 土震素B(TRB)的苯環部份.我們利用TMB 為半抗原,並接上載體蛋白KLH,合成免疫抗原,再將TMB-KLH以脾臟注射法注入紐西蘭 白兔中,引發TMB-KLH多株抗體.在利用TMB-BSA為吸附抗原,設立一酵素連結免疫吸附 分析系統,以競爭性間接ELISA進行交叉反應姓測試,發現TMB-KLH抗血清中存有可專一 性識別TMB之物質.在以此多株抗體為探針,並應用免疫組織化學技術與土麴菌俊體內之 TMB抗原反應並以ABC COMPLEX放大其訊號,在光學顯微鏡下觀察TMB及TRB在土麴菌23-1中 產生的部位.結果顯示TMB及TRB在早期產生於菌體之擔子柄,隨著菌體生長中期及後期,孢 子處也有免疫訊號,即可知孢子內也有TMB及TRB的生成. 3,4,5-Trimethoxy benzoic acid (TMB) is similar in structure of the aromatic moiety of terrutrem B (TRB), which is the secondary metabolite of Aspergillus terreus 23-1. The technique of immunomicroscopy with anti-TMB polyclonal antibody was applied to locate the site of TMB or TRB producing in the fungi during different period of cultivation. Anti-TMB serum was produced from New Zealand white rabbit by intra-spleen injection of the coupling of the haptenic TMB to keyhole limpt hemocyanin (KLH). Antigen-antibody complex were demonstrated by light microscopy using with immunohistochemical technique. The results indicated that TMB or TRB was formed in the sterigma of the fungi at the early stage of cultivation in potato dextrose media. However TMB or TRB was formed in the conidia at the middle and latter stage of cultivation.
