從腸炎弧菌 (Vibrio parahaemolyticus)中選殖出錳型超氧歧化(Mn-SOD) 之基因,並轉殖於表現載體pET-20b(+) ,轉型到大腸桿菌 (Escherichia coli) 表現宿主BL21(DE3)pLysS中進行蛋白質之大量表現。此一基因的開放讀架 (open reading frame) 含有588個核酸基因,可轉譯出196個氨基酸蛋白質,推算出其分子量為21713Da。利用金屬親和性管柱將大腸桿菌粗萃取物中的重組蛋白質純化出,並測試其生化特性。結果顯示,腸炎弧菌錳型超氧歧化重組蛋白質 (VPMn-SOD) 能夠耐受60℃的高溫,並對一些抑制物如:potassium cyanide (KCN), ethylenediaminetetraacetate (EDTA), sodium azide (NaN3) and diethyldithiocarbamic acid (DDC) 具有敏感性,而當sodium dodecyl sulfate (SDS) and hydrogen peroxide (H2O2) 存在時其活性會被抑制。其中心金屬離子經由graphite atomic absorption spectrophotometry (graphite AAS) and inductively coupled plasma (ICP) 等方法判定為錳離子。根據腸炎弧菌錳型超氧歧化基因所設計的專一性引子 (primer),以交叉聚合鏈 (Cross-PCR) 來建立一快速的方法,可用於弧菌屬之菌種鑑定。

The sodA gene coding for manganese superoxide dismutase (Mn-SOD) from the marine microorganism Vibrio parahaemolyticus was cloned, sequenced and over-expressed in Escherichia coli using the pET20b (+) expression vector. The full-length gene was consisted of 588 bp open reading frame and encoded a polypeptide of 196 amino acid residues, with a calculated molecular weight of 21713 Da. The recombinant enzyme was efficiently purified from crude E. coli cell lysate by the metal ion affinity chromatography. The recombinant VPMn-SOD resisted thermo-denaturation up to 60℃ and was insensitive to inhibitors such as potassium cyanide (KCN), ethylenediaminetetraacetate (EDTA), sodium azide (NaN3) and diethyldithiocarbamic acid ( DDC). The enzyme was inhibited by sodium dodecyl sulfate (SDS) and hydrogen peroxide (H2O2). The central metal ion of the recombinant VPMn-SOD was confirmed to be manganese by the methods of graphite atomic absorption spectrophotometry (graphite AAS) and inductively coupled plasma (ICP). The specificity of V. parahaemolyticus Mn-SOD gene probe was analyzed by cross-species PCR in order to provide the information for the purpose of Vibrio strain identification.
