本研究主要探討大蒜精油(garlic oil, GO)及其有機硫成分--二稀丙基硫化物 (diallyl sulfide, DAS)、二稀丙基二硫化物 (diallyl disulfide, DADS)及二稀丙基三硫化物 (diallyl trisulfide, DATS)調控Sprague-Dawley大白鼠免疫反應的活性。實驗以6~8週齡SD大鼠為對象,體外培養淋巴細胞,觀察大蒜精油及其有機硫成分DAS、DADS、DATS對淋巴細胞複製能力的效應;另外,觀察大蒜精油是否調控大鼠腹腔巨噬細胞在LPS活化下釋出NO的能力。結果顯示,淋巴細胞在75或250μg/ml ConA處理下,大蒜精油增加細胞增生的能力(p<0.05),且GO呈現劑量正相關的現象,當GO濃度為50μg/ml時有最大的效果; DAS和DADS組則增加了250μg/ml ConA處理組的淋巴細胞複製效果( p<0.05);然而,DATS對淋巴增殖能力則不具調控的效應。在腹腔巨噬細胞NO釋放能力分析中,結果顯示, 2、 10及50μg/ml的大蒜精油均可抑制活化型巨噬細胞釋出NO的能力,並呈現劑量關係的抑制效果( p<0.05);另外,也發現巨噬細胞形態改變、細胞蛋白質總量減少及胞外乳酸去氫活性增加。以上結果得知,大蒜精油及其有機硫成分DAS、DADS在免疫反應上可能扮演重要調控的角色。
