黑角舞蛾核多角體病毒株c782和榕樹透翅毒蛾核多角體病毒株具有不同之體外寄主域。c782病毒株只能感染舞毒蛾細胞株,榕樹透翅毒蛾核多角體病毒株則對舞毒蛾細胞株及榕樹透翅毒蛾細胞株皆可感染,並呈現多角體病變。本論文分別由此二病毒株各自分別感染舞毒蛾和榕樹透翅毒蛾細胞株後產生之子病毒的效價、吸附係數和螢光抗體偵測,評估二病毒株各在兩種細胞內之表現,證實c782病毒株不能在榕樹透翅毒蛾細胞株內複製。從二病毒株所建立的基因庫中選殖和定位此二病毒之 gp64 和 hel基因,並與已發表的種類作比較其相似度,以初步判定其所扮演之角色。此外,依據基因序列與已發表的相同基因序列所繪出之親緣分支樹,可為其演化過程親緣遠近之參考。本論文排除了gp64是影響寄主域之主因,至於hel之角色和hel與其他基因在寄主域上之互動仍有待進一步之研究。
Two viruses, Lymentria xylina nucleopolyhedrovirus c782 strain (LyxyNPV c782) and Perina nuda NPV θ strain (PenuNPVθ), exhibited in vitro host range divergence in IPLB-LD-652Y and NTU-PN-HH cells. Both LD and PN cells were susceptible toθvirus and showed a typical cytopathogenesis of polyhedrosis, while only LD cells were susceptible to c782 virus. The infectivity of c782 orθ virus, including the adsorption efficiency, the viral titers of the progeny viruses at 7-day postinfection, and immnofluorescent assay at 1- to 5-day postinfections, were examined. The results led to a conclusion that c782 virus can invade PN cells but not replicate in PN cells. The plasmids containing gp64 and hel genes were selected from BamHI and EcoRI gene libraries, respectively, that were established from LyxyNPV or PenuNPV. The localization of both genes in the restriction enzyme fragments of LyxyNPV or PenuNPV DNA was determined. The complete nucleotide sequences of both genes of LyxyNPV or PenuNPV were carried out. The comparison of gp64 or hel gene sequences of two NPVs with known gp64 or hel gene sequences from genbank was also done. Based on the sequences of both genes, the phylogenetic trees were constructed. Although we had confirmed that gp64 is not a key factor on the infectivity of c782, the real role of hel or the relationship of hel with other association factors (or genes) on their host range still need to be elucidated in the further studies.
