本研究首先利用美國國家衛生院(National institutes of health; NIH)建立之NCBI(National center for biotechnology information)網站,搜尋到許多可能是鋅指蛋白基因之EST(expressed sequence tag)clone ,再依據各EST clone之核酸序列設計適當引子對(primers pairs),並針對源自小鼠各不同器官組織所萃取之總RNA(total RNA),進行反轉錄聚合鍊鎖反應(reverse transcription-polymerase chain reaction ;RT-PCR),從而比較小鼠各器官組織中鋅指蛋白基因之表現情形。試驗結果發現,zfp 553在腦、心臟、腎臟等組織僅有微量之表現;而在睪丸及子宮等組織中,其表現量則高於前述者。此外,應用北方雜交反應(Northern hybridization)技術,初步證明該基因全長約為2.5 Kb;進一步針對另一具有該基因全長cDNA之EST clone進行定序結果,更確認不含poly A tail 之cDNA,其全長為2308 bp。當以MacVectorTM軟體針對該cDNA序列進行分析,結果發現此一基因係可以轉譯出一個具有478個胺基酸、分子量約為52KDa、包含有3個Cys2-His2鋅指結構之鋅指蛋白。
鑑於RT-PCR及北方雜交反應結果顯示,zfp553基因在子宮表現量高於其他組織者,且其表現量將隨懷孕期之進展而有改變;此外,應用原位雜交反應(in situ hybridization)技術測試結果,更證實該基因之表現位置主要是在子宮內膜及腺體之上皮細胞,因此似可推論zfp553基因之作用,應涉及維持小鼠懷孕所需之子宮環境的基因調控機制。
To identify novel zinc finger proteins expressed in the mouse uterus during pregnancy, we first searched the mouse expressed sequence tag(EST)data bases for the putative zinc finger genes with conserved zinc finger domain. Several novel zinc finger containing genes were identified with this approach. We then examined the expression patterns of these genes during different stages of pregnancy as well as different tissues by reverse transcrption-polymerase chain reaction(RT-PCR)analysis. A novel zinc finger gene, zfp553, which is differentially expressed in uterus during different stages of pregnancy was identified. The full length of zfp553 cDNA is 2308-bp which contains an open reading frame encoded 478 amino acids. The deduced amino acid sequence of zfp553 contains three Cys2-His2-type zinc finger motifs. In situ hybridization analysis revealed that zfp553 is expressed predominantly in the uterine epithelial cells. According to these results, the zfp553 may play an important role in maintaining uterine environment during pregnancy.
