摘要
高山白腹鼠(Niviventer culturatus)及台灣森鼠(Apodemus semotus)是兩種台灣地區的特有種嚙齒類動物。這兩種鼠類廣泛分布在台灣中央山脈海拔1,500公尺以上的山區,是台灣許多山區小型哺乳類的優勢物種。主要棲息在闊葉林、針闊葉林及針葉林的底層灌叢,但台灣森鼠也經常在玉山箭竹草原區出沒活動。
本研究利用PCR-RFLP的方法分析臺灣地區20個族群,128隻高山白腹鼠及23個族群、271隻台灣森鼠的地理類緣關係。利用聚合連鎖反應(PCR)分別增幅高山白腹鼠粒線體DNA之CYTb/DL(約2,800 bp)及12S/16S(約1,900 bp)及台灣森鼠粒線體DNA之CYTb/DL基因片段,再分別以13種及11種限制內切來分析這兩種鼠類之增幅片段的限制片段長度多型性(RFLP)。其中高山白腹鼠共得到25個粒線體DNA的單倍基因型(haplotype)。各單倍基因型的序列差異介於0.32%至2.63%之間,族群內粒線體DNA的分歧度估計值介於0%至0.69%之間,而族群與族群間的差異介於-0.08%至1.71%之間。除了少數幾個單倍基因型的族群地理分布較廣之外,大多數的單倍基因型都只發現在單一個族群或相鄰近的族群內,各單倍基因型之間並未發現明顯的地理類緣關係組合。另外,在台灣森鼠部分,共獲得44個粒線體DNA的單倍基因型,各單倍基因型的序列差異介於0.20%至3.20%之間。透過各單倍基因型之間的類緣關係分析,可以發現四個主要的遺傳分枝群,各遺傳分枝群的序列差異介於0.86%至1.78%之間。雖然台灣森鼠這四個遺傳分枝群的基因歧異度和地理分布頗為複雜,但仍可發現各遺傳分枝群都侷限分布在由北至南的特定地理區域中。藉由七個主要單倍基因型的地理分布及各族群UPGMA的分析,本研究將所有的森鼠族群區分成北部及中南部山區等兩個地理分群。
高山白腹鼠及台灣森鼠這兩種棲息在台灣高海拔山區的鼠類,雖然棲息分布的區域極為相近,但族群間的類緣關係卻不相同。這可能是因為受到不同歷史地質事件(vicariance)及不同族群擴散(dispersal)過程的影響。
