在本研究中我們利用McMurry的合成方法以Ti-Zn為反應試劑,且以泰母芬(Tamoxifen)為骨架,改變苯環上之取代基與烷基修飾其結構,而製備1-(4-胺苯基)-1,2-二苯基-1-丁烯;1-(4-胺苯基)-1,2-二苯基-1-丙烯;1-(4-胺苯基),1-(4-氟苯基)-2-苯基-1-丁烯以及1-(4-胺苯基),1-(4-氟苯基)-2-苯基-1-丙烯,及在碘的摧化下與丙酮反應,製備含新型二甲基喏(dimethyl quinoline)取代基之1-(2,4-二甲基喏林)-1,2-二苯基-1-丁烯;1-(2,4-二甲基喏林)-1,2-二苯基-1-丙烯;1-(2,4-二甲基喏林)-1-(4-氟苯基)-2-苯基-1-丁烯及1-(2,4-二甲基喏林)-1-(4-氟苯基)-2-苯基-1-丙烯,並將這些合成之產物做抗癌活性及藥理活性之評估。

The Tamoxifen skeleton with various substituents were prepared by using a McMurry's method with titanium-zinc system as coupling reagent. In this work,1-(4-aminophenyl)-1,2-diphenyl-1-butene;1-(4-ami-nophenyl)-1,2-diphenyl-1-propene;1-(4-aminophenyl),
1-(4-flurophenyl)-2-phenyl-1-propene;1-(4-aminophenyl),1- (4-flurophenyl)-2-phenyl-1-butene were synthesized under this conditions.
Above results were reacted with a doubly nucleophilic reagent, acetone and anilines with the presence of iodine as a catalysis, to yield 1-(2,4- dimeythylquinoline)-1,2-diphenyl-1-propene;1-(2,4-dimeythylquinoline)-1,2-diphenyl-1-butene.
All the compounds have prepared for cytotoxic test, anticancer activity, and pharmacological activity.
