本研究的目的是應用數量遺傳與分生技術,探討臺灣地區飼養之黑色豬隻之遺傳特性。本研究共分成三章進行,第一章膚色及重要經濟性狀之遺傳特性,第一節調查桃園種 (Taoyuan, T) 與杜洛克 (Duroc, D) 各種配種型態產生後代之膚色分離情形,並以矯正之卡方測定進行遺傳分離適合度檢定,以瞭解桃園種全黑色膚色的基因與杜洛克全紅色膚色基因有無顯隱性關係。第二節應用台灣黑豬選育初期進行之T與D進行正反交配,所得四種配種型式 (D ×D、D ×T、T ×T及T ×D)共72胎豬隻的繁殖性能及後裔豬隻的生長性能、體型及屠體資料,並應用SAS之PROC Mixed分析軟體及Malik (1984) 遺傳效應估算方式,進行T與D豬隻的相對遺傳效應估算。第二章基因組DNA指紋分析,第一節應用豬基因組DNA的逢機增殖多態性DNA ( Random Amplified Polymorphic DNA, RAPD) 指紋比較D及T之遺傳差異,並進一步評估此遺傳差異作為T與D雜交育成豬種之血緣關係分析的可行性。第二節應用 OPAA、OPAO、OPAV、OPC及OPE等系列 Operon 10 mer 逢機寡核酸引子及分生技術,去增殖不同品種豬隻基因組DNA,探討是否可增殖多態性的RAPD指紋後,選其中可產生多態性環帶的OPAV05 與OPAV15二種逢機引子,對不同品種豬隻與來自臺灣各地區黑色豬隻基因組DNA進行逢機增殖,所形成RAPD多態性指紋,計算其共有環帶頻率、不同品種間相似性及遺傳距離,進行臺灣各地區飼養的黑色豬隻之遺傳特性及類緣關係分析。第三章粒腺體DNA D-loop分析,第一節依據豬粒腺體DNA D-loop區域已知序列,設計一組可以豬之基因組DNA為模板,經由聚合連鎖反應,增殖出D-loop區域392-bp片段的引子,利用此組引子增殖不同品種豬隻之D-loop區域392 bp片段,進行臺灣各品種豬隻粒腺體DNA D-loop區域之單股構形多態性 ( Single Strand Conformation Polymorphism,SSCP) 分析,並探討依據母系血緣推定之梅山母豬粒腺體DNA D-loop區域SSCP態樣與其繁殖性狀之相關性。第二節利用上述設計之引子增殖不同品種豬隻及臺灣各地區黑色豬隻之粒腺體DNAD-loop區域392 bp片段,進行單股構形多態性,以了解臺灣各地區黑色豬粒腺體DNA D-loop區域差異性及其類緣關係分析。
第一章膚色及重要經濟性狀之遺傳特性,試驗結果顯示: 1. T的全黑色基因對D全紅色基因顯性。此兩豬隻品種若進行雜交,可經選拔育成TB黑色豬隻。 2. 雜種後裔D-T雜種傾向比D品種有較好平均窩仔數和三週齡活頭數,比T品種有較好的生長性狀、體型及屠體性狀。第二章基因組DNA指紋分析,試驗結果顯示:1. T品種個體基因組DNA與歐美品種D個體不同。 2. 畜試黑豬一號 ( Taiwan Livestock Research Institute Black Pig No. 1, TB)遺傳相似性達74.5%,而其種原品種 D 為74.7%,T為74.2%,再經由遺傳距離分析得知畜試黑豬遺傳距離較接近D,這提供了RAPD指紋可作為決定T、D及畜試黑豬一號間血緣關係的例證。 3. 所使用的OPAA、OPAO、OPAV、OPC及OPE等系列引子共100種中,有58種引子經RAPD-PCR增殖後可產生多態性的環帶。 4. 各參試豬隻品種內共有環帶頻率介於0.753-0.909之間。峨嵋與關廟兩地區黑色豬與TB 比其他地區有較高的遺傳相似性 (0.894與0.876)。5.若以T、D、漢布夏及盤克夏四個品種與各地區黑色豬進行相似性分析,嘉義、彰化、三峽及樹林黑色豬各分別與T相似性較高,而關廟、淡水及峨嵋黑色豬則各分別與D相似性較高。第三章粒腺體DNA D-loop分析,試驗結果顯示: 1. 歐美品種豬隻的SSCP電泳態樣皆有A 態樣,而亞洲品種豬隻的SSCP電泳態樣則無A態樣。歐美品種豬隻的SSCP電泳態樣除約克夏及漢布夏外,餘皆無C態樣,C和D態樣存在於亞洲品種豬隻居多。 2. 粒腺體DNA D-loop區域之SSCP態樣有潛力成為選拔梅山豬仔豬出生後早期生長速率的有用標誌。 3. 所有參試黑色豬單股構形多態性態樣分佈為75 A ( 30.6%), 47 B ( 19.2%), 31 C ( 12.6%) 和 92 D ( 37.6%)態樣。畜試所TB有A、B和D態樣與其他地區黑色豬的態樣分佈有不同。 4. 由粒線體DNA D-loop區域SSCP分析結果,推測關廟黑豬的雌親物質來自歐美品種,而彰化黑豬來自亞洲品種。
