本研究主要比較大蒜精油 (garlic oil, GO)及其有機硫化物─二丙烯基硫化物 (diallyl sulfide, DAS)、二烯丙基二硫化物 (diallyl disulfide, DADS)及二烯丙基三硫化物 (diallyl trislfide, DATS)調控巨噬細胞 iNOS表現之活性。在0.1 g/ml脂多醣體 (Lipopolysaccharides, LPS)誘發巨噬細胞RAW 264.7條件下,不同濃度GO (36.5g/ml)、DAS (100、300M)、DADS (100、300M)、DATS (50、100M)的培養並不影響細胞存活率,且DADS (100、300 M)及DATS (50、100M )顯著抑制巨噬細胞nitric oxide的釋放能力 (P<0.05),其中又以DATS抑制效果最強,DAS則無效果;蛋白質分析 (Western blotting) 結果顯示當細胞以DADS或DATS處理時,iNOS蛋白質含量明顯低於對照組 (+DMSO),且呈現劑量效應,DATS抑制iNOS蛋白質效果最強,至於IκB含量也以DADS 及DATS處理組較DAS高;Northern blotting所得iNOS mRNA表現與蛋白質分析結果一致。綜合以上結果,GO抑制iNOS表現效應與精油中的DADS及DATS有關,尤其DATS的角色最重要,抑制巨噬細胞iNOS的表現推論應與IκB/NFκB途徑有關。
