稻穀貯藏是一老化過程。老化是一氧化逆境,造成生物體氧化傷害。生物體為了避免遭受氧化傷害,發展出一保護機制用以清除自由基與過氧化物,包括抗氧化清除酵素與抗氧化物。本試驗為了評估抗氧化酵素在種子貯藏期間的變化,以台稉8號、台稉9號、台中秈10號、台中189號、高雄142號及台農67 號的糙米為試驗材料,貯藏在15 ℃、25 ℃及35 ℃,相對濕度40 %的環境下,調查抗氧化清除酵素包括觸酶、過氧化酶、超氧歧化酶、榖胱苷肽還原酶、抗壞血酸過氧化酶、脫氫抗壞血酸過氧化酶及榖胱苷肽過氧化酶等活性的變化。
試驗結果顯示,隨著稻穀貯藏期間,穎果中七種抗氧化酵素的活性均隨之降低。胚中SOD、POD、APX及GR活性與穎果相同,皆隨貯藏時間增加活性降低,其中以35 ℃貯藏者下降較多,25 ℃貯藏者次之,15 ℃貯藏者下降較緩慢。唯胚中CAT、GPX及DHAR在貯藏溫度之間差異較不明顯。
這些酵素活性在貯藏期間,隨貯藏時間增加活性下降的趨勢,六個參試品種表現皆相同,此結果顯示抗氧化酵素活性的變化,可作為判斷糙米貯藏新鮮程度的指標。
另外利用POD活性於貯藏期間下降的趨勢,進一步以benzidine及dianisidine為反應物偵測POD活性,以光電比色法讀取吸光值,可以判斷POD活性,呈色反應液的光電比色法除可以用於混和米樣POD活性檢定外,亦可以適用單粒米之間POD活性的檢定。利用POD呈色的光電比色法,以建立區別不同期作稻米混和比粒的檢驗方法。
