假性狂犬病毒(Pseudorabies virus, PRV)、豬小病毒(Porcine parvovirus, PPV)、豬環狀病毒(Porcine circovirus, PCV)、豬瘟病毒(Classical swine fever virus, CSFV)、Porcine reproductive and respiratory syndrome virus(PRRSV) 及日本腦炎病毒(Japanese encephalitis virus, JEV)乃是豬隻在臨床上常見的病毒性感染。我們遂針對本省豬隻重要病毒性疾病之病原進行的相關研究,目前已完成部分病毒基因的選殖和核酸序列分析,可提供作為研究生物晶片的材料。而本實驗亦已建立一項可分別針對假性狂犬病毒之gE、gX基因與豬小病毒NS-1基因及豬環狀病毒第一型之ORF1基因、第二型之ORF2基因等病毒基因的多引子聚合酶連鎖反應(multiplex PCR)快速診斷方法。此外,我們亦針對豬瘟病毒之5’UTR、NS5基因與PRRSV之ORF3基因及JEV之NS5等基因開發多引子反轉錄聚合酶連鎖反應(multiplex RT- PCR)之快速診斷方法持續應用此一技術作為基礎,進而發展生物晶片技術以快速且特異性地診斷鑑定多種豬隻重要之傳染性疾病。
Pseudorabies virus (PRV), porcine parvovirus (PPV), porcine circovirus (PCV), classical swine fever virus (CSFV), porcine reproductive and respiratory syndrome virus(PRRSV)and Japanese encephalitis virus (JEV) are the most commonly occurrence of virus infections in pig populations in Taiwan. We have studied on those important swine viruses and several viral target genes were cloned and sequence analysis for further application on the microarray studies. The specific PCR primer pairs for each viral target gene were designed and the specifity as well as sensitivity of each primer were also determined. A multiplex PCR technique using the primer pairs specific to the gE and gG genes of PRV, NS1 gene of PPV, ORF1 gene of PCV1 and ORF2 gene of PCV2 was successfully set up for rapid diagnosis of PRV, PPV and PCV in clinical specimens. In addition, a multiplex RT-PCR was also developed to detect target the genes including 5’UTR and NS5 genes of CSFV, ORF3 gene of PRRSV and NS5 gene of JEV.
