因缺血性缺氧所造成的腦部傷害可能由於腦神經細胞死亡所產生。經由高壓氧治療可以增加溶解於血漿中氧氣含量,因此當暫時性腦缺血事件發生後接受早期高壓氧治療證實可以改善傷害的程度,但高壓氧治療在劑量及方式上是一個重要問題。高壓氧治療機轉可能包含增加周圍細胞供氧量及缺血週邊氧量。先前一些文獻報告高壓氧治療可以減少腦部梗塞面積,腦梗塞機率及缺血性中風運動症狀缺損,然而卻有學者提出晚期治療其結果是有害的。目前高壓氧治療對於細胞層次的機轉仍然不清楚。本實驗利用中大腦動脈阻塞方式,造成大白鼠缺血性中風後,將大白鼠放入高壓氧艙中接受治療,然後先評估大白鼠行為症狀,接著用TTC染色來觀察其腦梗塞面積變化,再利用TUNEL染色來觀察神經細胞凋亡的情形。實驗結果在接受早期高壓氧治療後之大白鼠其大腦皮質有降低細胞凋亡數目,顯示早期高壓氧治療可能有助於增加缺血性中風大白鼠神經細胞存活。

Hypoxic-ischemic brain injury occurred could lead to brain cell death. To increase the amount of oxygen dissolved in blood plasma is by means of hyperbaric oxygen (HBO). HBO can improve the outcome after temporary focal ischemia when the treatment is started early after ischemia. Potential mechanisms of HBO treatment include enhanced oxygen supply to marginally perfused cells and ischemic periphery. Several reports in the literature had shown that hyperbaric oxygen reduced cerebral infarction area in animals. Recent reports showed the HBO therapy can decrease the incidence of infarction and attenuate motor deficits. So we want to know the mechanism of the HBO treatment. We applied the middle cerebral artery (MCA) occlusion model, then placed the rats in the HBO chamber according to the protocol. The rats were evaluated with behavioral neurological test and cerebral infarction area. The apoptosis was also examined with TUNEL stain and counterstained with DAPI. Our data suggest that the early treatment of HBO decrease the apoptotic cells after postischemic stroke in rats. So the early treatment of HBO may enhance the neuron survival after postischemic stroke in rats.
