摘要
以微透析裝置連結高效能液相層析儀分析牛奶中的六種磺胺劑,磺胺嘧啶、磺胺一甲基嘧啶、磺胺二甲基嘧啶、磺胺噁唑、磺胺一甲氧嘧啶、磺胺二甲氧嘧啶的含量,就HPLC系統和微透析系統進行多項變因的探討。層析條件包括動相和層析管柱的選擇;透析條件包括樣品溶液的pH值和添加鹽類濃度、灌注液添加極性修飾劑比例、灌注液流速和pH值及收集時間等影響透析之因素,找出最佳條件。
實驗結果以超純水為灌注液,流速為1.06 μl/min,收集20分鐘條件下,連線HPLC動相為75﹪,pH 5.0,10-2M磷酸二氫鈉緩衝溶液混合25﹪氫甲烷,搭配逆相C-8管柱進行沖提,分析主要六種磺胺劑。
在最佳層析及微透析條件下分析得回收率為SDA:305.99﹪、SME:284.18﹪、SMX:177.89﹪、SMZ:452.35﹪、SMM:268.72﹪、SMO:482.94﹪;偵測極限為SDA:0.151ppb、SME:0.130ppb、SMX:0.038ppb、SMZ:0.002ppb、SMM:0.002ppb、SMO:0.003ppb。研究並針對市售牛奶進行磺胺劑含量的檢測。

Abstract
The purpose of the study was to analyze 6 sulfonamides, namely Sulfadiazine (SDA), Sulfamerazine (SME), Sulfamethazine (SMZ), Sulfamethoxazole (SMX), Sulfamonomethoxine (SMM), Sulfadimethoxine (SMO), in milks by micro-dialysis connected with HPLC. The conditions of HPLC analysis included the stationary phase selection and the mobile phase was examined. Also the conditions of micro- dialysis that included the pH and the concentration of salt adding of the sample as well as the pH, flow rate, and the modifier of perfusate were examined.
The optimal conditions of the micro-dialysis sampling were pure water as perfusate with the flow rate of 1.06 L/min and 20 min. collection time. And the optimal conditions of HPLC analysis were C8 reversed phase column with the mobile phase of 25﹪acetonitrile and 75﹪, pH 5, 10-2 M phosphate buffer.
By the optimal conditions, the recoveries of micro-dialysis were 305.99%, 284.18%, 177.89%, 452.35%, 268.72%, and 482.94% for SDA, SME, SMX, SMZ, SMM and SMO, respectively. And the detection limits of 0.151, 0.130, 0.038, 0.002, 0.002 and 0.003 ppb were found for SDA, SME, SMX, SMZ, SMM and SMO, respectively for the system of micro- dialysis connected with HPLC .The study also examined the commercial milks by the method and the results are reported.
