摘要
以微透析裝置連結高效能液相層析儀分析果汁中的四種有機酸,即酒石酸、蘋果酸、乳酸、檸檬酸的含量,就HPLC系統和微透析系統進行多項變因的探討。層析條件包括動相和層析管柱的選擇;透析條件包括樣品溶液的pH值和添加鹽類濃度、灌注液添加極性修飾劑比例、灌注液流速和pH值及收集時間等影響透析之因素,找出最佳條件。
實驗結果以超純水為灌注液,流速為1.06 μl/min,收集15分鐘條件下,連線HPLC動相為pH2.0,5×10-2M磷酸緩衝溶液,搭配逆相C-18管柱進行沖提,分析主要四種有機酸。
在最佳層析及微透析條件下分析得偵測極限為TA:434.1735ppb、MA:831.6650ppb、LA:1577.8417ppb、CA:1426.7846ppb。研究並針對市售果汁進行有機酸含量的檢測。

Abstract
The purpose of the study was to analyze 4 organic acids, namely tartaric acid(TA), malic acid(MA), lactic acid(LA), citric acid(CA), in juice drinks by micro-dialysis connected with HPLC. The conditions of HPLC analysis included the stationary phase selection and the mobile phase was examined. Also the conditions of micro-dialysis that included the pH and the concentration of salt adding of the sample as well as the pH, flow rate, and the modifier of perfusate were examined.
The optimal conditions of the micro-dialysis sampling were pure water as perfusate with the flow rate of 1.06 mL/min and 15 min. collection time. And the optimal conditions of HPLC analysis were C18 reversed phase column with the mobile phase of pH=2, 5×10-2 M phosphate buffer.
By the optimal conditions, the detection limits of micro-dialysis were 434.1735ppb, 831.6650ppb, 1577.8417ppb, and 1426.7846ppb for TA, MA, LA and CA, respectively for the system of micro- dialysis connected with HPLC .The study also examined the commercial juice drinks by the method and the results are reported.
