游離酵素因為反應時環境因子的變化，如溫度、pH值等，而導致活性的喪失，所以使用一次後即需丟棄，相當浪費。經固定化後的酵素不僅對於環境變化穩定性高，且因為固定於擔體上的緣故，使其在回收再利用上變的相當方便。不過雖然固定化酵素有許多優點，但是酵素固定化的效率卻常常不甚理想，因此藉由探討酵素固定化反應的最適條件來提升固定化的效率，是酵素固定化研究中相當重要的一環。
本研究係將Candida rugosa Type Ⅶ 脂肪分解酵素在液相中分別以交聯(cross-linking)及吸附(absorption)兩種方式固定在幾丁聚醣顆粒上。本研究所使用之交聯劑為戊二醛(Glutaraldehyde)。在交聯方面，經實驗後發現，在pH 7.0下，使用0.0025 %（v/v）之Glutaraldehyde和幾丁聚醣進行預先交聯30分鐘後，再使用1%（w/v）之脂肪分解酵素進行固定化反應1小時，可得較佳之固定化比活性30.33 U/mg-protein，其固定化效率為40.3%。吸附方面，在pH9.0下，以1%（w/v）之脂肪分解酵素進行固定化反應2小時，可得較佳之固定化比活性 54.62 U/mg-protein，其固定化效率為64%。在生化性質性質探討方面，水解反應最適pH值由游離酵素的pH9.0變動為pH8.0；水解反應最適溫度則提高至50℃。經固定化後的酵素其pH安定性可由原來游離酵素的pH在5~7延長至pH在5~8；而溫度安定性可由原來游離酵素之40℃以內增加到50℃。當使用批次方式來進行水解反應時，以吸附法所製備之固定化酵素在經過連續10次的回收後，其比活性僅存20%；但以交聯法所製備之固定化酵素回收10次後卻仍可保有64 %以上之比活性。
