本篇論文報導氫咪唑苯二氮雜草銅類似物之合成. 在以1,4-dioxane為溶劑且醋酸鈉溶液(0.3 M)為緩衝溶液的環境下, 經由溴化反應使6-amino-9-(2-chloroethoxymethyl)purine (6) 反應生成8-bromo-9-(2-chloroethoxymethyl)adenine (10). 化合物10可進行親核基取代反應,使氮原子被碘原子取代進而生成化合物12. 此外, 化合物10亦可與甲醇或n-Bu4NOH進行反應, 分別生成其衍生物13或15.
Synthesis of a new tetrahydrobenzodiazepinone analogs as anti-HIV agent was accomplished. First, 8-bromo-9-(2-chloroethoxymethyl)adenine (10) was prepared by treatment of 6-amino-9-(2-chloroethoxymethyl)purine (7) with bromine in NaOAc buffer solution (0.30 M) and 1,4-dioxane. Nucleophilic substitution of the chlorine atom in 10 produced iodo derivative 12. Reaction of 10 with methoxy or hydroxy group gave derivatives 13 and 15, respectively. Cyclization of compound 13 was performed by addition of catalytic amout of (NH4)2SO4 in hexamethyldisilazane.
