摘要
由於二十二碳六烯酸(DHA),為人體大腦及視網膜神經組織中結構脂質的成份,扮演著重要的角色,為有價值的營養物質。此外DHA和其他ω-3系列脂肪酸,為預防人類各種疾病的重要物質。魚肉通常為日常飲食中這些脂肪酸的來源,而魚類這些脂肪酸的起源為來自於餌料中的海洋微生物。因而這些海洋微生物可於控制之下大量培養,成為取代這些ω-3系列脂肪酸之來源。
海洋單細胞微藻Ischrysis galbana CCMP 1324含豐富的DHA,並且早已廣泛應用於水產養殖當作生物餌料來源,於DHA生產上具有很大的潛力。本實驗利用批次式培養與修飾Walen’s培養基,探討Ischrysis galbana CCMP 1324的營養源攝取與DHA的生產情形。結果顯示,於培養的第八天,利用修飾培養基所生產的細胞濃度為Walen’s培養基的1.03倍。在DHA累積方面,於對數生長期斷除氮源並添加有機碳醋酸鈉進行不照光培養,則DHA的產量佔乾重的1.8﹪(中性DHA:1.26﹪,極性DHA:0.54﹪),為Walen’s培養基的1.58倍。
Abstract
Docosahexaenoic acid (DHA)is valuable nutrient because it has an important role as the constituent of structural lipids in the nervous tissue of human brain and retina. In addition, DHA and other ω-3 fatty acids are important factors in the prevention of several human diseases. Fishes have been the dietary source of these fatty acids and they originate from the marine microbes upon which the fishes feed. Therefore, these marine microbes cultured largely under control could be utilized as an alternative source of the ω-3 fatty acids.
The rich in DHA marine unicellular alga, Isochrysis galbana CCMP1324, has been used as a living food source for aquaculture and has great potential for DHA production. This study was to investigate the nutrient uptake and DHA production of Isochrysis galbana CCMP1324 by using batch culture and modified Walne’s media.The results showed that the cellular density of this species culturing with modified media was 1.03 times higher than that culturing with Walne’s media at eighth day. As for the DHA accumulation of biomass, Isochrysis galbana CCMP1324, it has 1.8 %(neutral 1.26%, polar 0.54%)of dry weight about 1.58 times higher than the Walne’s media culture. When this alga at log phase then was continually cultured at dark room under a non-nitrogen source and offering sodium acetate in media.
