在本論文中首先利用靈芝甲醇萃取物為材料,對不同的癌細胞株進行毒性試驗,結果發現靈芝甲醇萃取物能夠有效抑制腫瘤細胞株生長,包括有肝癌細胞株 (HuH-7)、大腸直腸癌細胞株 (HCT-116) 、Epstein Barr virus所轉型之B淋巴細胞 (Raji) 、血癌細胞株 (HL-60),其中以抑制肝癌細胞株生長之能力最強。
由靈芝甲醇萃取物中利用兩次矽膠管柱層析法,並配合細胞毒性試驗分析,分離並純化出抗癌活性成分。結果顯示,M7-4 fraction對HuH-7比靈芝甲醇粗萃液有更強的毒殺效果,經24小時處理對HuH-7細胞IC50為40 mg/ml,我們還進一步發現M7-4對HuH-7的毒殺作用是經由透過粒腺體參與的細胞凋亡路徑。我們檢視了與細胞凋亡相關的蛋白,結果發現用M7-4處理HuH-7細胞後,能增加促進凋亡分子Bcl-xS、caspase、MAPKs的活化,以及降低抗凋亡分子Bcl-xL,使細胞走向凋亡。在調控細胞生長相關分子的研究上,我們發現靈芝三帖類能使HuH-7細胞中PKC的活性下降,並降低其下游分子c-myc的表現量。
由上述的結果得知,靈芝三帖類能經由抑制細胞的生長、增加促凋亡蛋白表現誘導HuH-7細胞走向凋亡。此結果有助於以後從靈芝發展成抗癌藥物。
