將乾燥棘蟻15.0 kg以乙醇迴流萃取,所得之萃取液經減壓濃縮後,分別以正己烷、氯仿、乙酸乙酯、正丁醇與水進行多次分配萃取,各分配層以管柱層析、薄層層析、及再結晶方式處理,共得到49個化合物,其中43個經光譜分析確認其化學結構為:lamellidine-A、lamellidine-B、lamellidine-C、lamellidine-D、lamellidine-E、lamellidine-F、lamellidine-G、3-isobutyl-6-methyl-2,5-diketopiperazine、N-valyl-tyrosine、 N-leucyl-tyrosine、N-leucyl-tryptophan、L-tyrosine、L-leucine、L-isoleucine、L-alanine、 L-glutamic acid、L-arginine、L-tryptophan、L-phenylalanine、L-3-(3,4-dihydroxyphenyl)- alanine、3-p-hydroxyphenyl-3-amino-1-propanoic acid、β-carbolin-1-ol、β-carboline、stellarboline-N、stellarboline-V、tussilagoline-A、tussilagoline-B、nicotinic acid、thymine、uracil、transtorine、quinoline-3-carboxaldehyde、adenosine、quinolinic acid、indole-3-carbaldehyde、6-biopterin、xanthopterin、protocatechuic acid、4-hydroxybenzoic acid、piperonal 、cholesterol、cholest-4-en-3-one、hydroxyecdysone。
其中,lamellidine-A、lamellidine-B、lamellidine-C、lamellidine-D、lamellidine-E、lamellidine-F、lamellidine-G、N-valyl-tyrosine和β-carbolin-1-ol等9個為天然界首次分離得到的新化合物。
The dried black ants ( Polyrhachis lamellidens ) 15.0 kg were extracted with EtOH, and then concentrated under reduce pressure to obtain dark brown crude extract 1.4 kg. The crude extract was partitioned between H2O with n-hexane, and then with CHCl3, EtOAc and n-BuOH. Totally, forty-nine compounds were isolated by chromatographic methods in this study. The structures of forty-three compounds had been determined by spectral analysis. They were lamellidine-A, lamellidine-B, lamellidine-C, lamellidine-D, lamellidine-E, lamellidine-F, lamellidine-G, 3-isobutyl-6-methyl-2,5-diketopiperazine, N-valyl-tyrosine, N-leucyl-tyrosine, N-leucyl-tryptophan, L-tyrosine, L-leucine, L-isoleucine, L-alanine, L-glutamic acid, L-arginine, L-tryptophan, L-phenylalanine, L-3-(3,4-dihydroxyphenyl)-alanine, 3-p-hydroxyphenyl-3-amino-1-propanoic acid, β-carbolin-1-ol, β-carboline, stellarboline-N, stellarboline-V, tussilagoline-A, tussilagoline-B, nicotinic acid, thymine, uracil, transtorine, quinoline-3-carboxaldehyde, adenosine, quinolinic acid, indole-3-carbaldehyde, 6-biopterin, xanthopterin, protocatechuic acid, 4-hydroxybenzoic acid, piperonal, cholesterol, cholest-4-en-3-one and hydroxyecdysone.
Among them, lamellidine-A, lamellidine-B, lamellidine-C, lamellidine-D, lamellidine-E、lamellidine-F, lamellidine-G, N-valyl-tyrosine and β-carbolin-1-ol were first isolated from the natural source.
