中文摘要
本研究之目的在於建立簡單、快速、準確的高效液相層析法,以檢測雞飼料中馬杜拉黴素(maduramicin)與沙利黴素(salinomycin)。實驗中添加馬杜拉黴素與沙利黴素於自配空白雞飼料,經測試各種不同方法後,以乙腈萃取飼料中藥物,然後分別以dansylhydrazine和2,4-dinitrophenylhydrazine將藥物衍生化後注入HPLC系統檢測。馬杜拉黴素的分析條件如下:Symmetry® column(C18, 5μm; 3.9x150 mm I.D.);移動相:acetonitrile:6.6 mM tetrabutylammonium hydrogen sulfate(85:15, v/v),流速:1.0 mL/min,注入量:20 µL,螢光檢測器之激發波長:237 nm,放光波長: 500 nm。實驗結果顯示,馬杜拉黴素標準品滯留時間為17分鐘,最低檢測極限為1.6 ng(3.3σ/S),而最低定量極限為4.8 ng(10σ/S)。空白飼料中添加馬杜拉黴素(2.5~15 ppm),所得回收率為96~107 %,並成良好之線性關係,r2=0.994。沙利黴素的分析條件如下:Symmetry® column(C18, 5μm; 3.9x150 mm I.D.);移動相:methanol:1.5%(v/v)acetic acid(pH 3.0)(95:5, v/v),流速:1.0 mL/min,注入檢體量:20µL,紫外光檢測器定量波長:392 nm。實驗結果顯示,沙利黴素標準品滯留時間為5.4分鐘,最低檢測極限為3.5 ng(S/N³3),而最低定量極限為10.5 ng(S/N³10)。空白飼料中添加沙利黴素(15~120 ppm),所得回收率為102.1~112.3 %,並成良好之線性關係,r2=0.999。
