Uroteuthis (Photololigo) chinensis、U. (P.) edulis與U. (P.) duvaucelii是棲息環境重疊且外在形態非常相似的物種。三物種的分類特徵有吸盤角質環形態、鰭長比例、莖化腕莖化長度比例等,但未成熟個體的分類特徵尚未發育完全,因此更加難以分類鑑定,而無法建立生物的基礎資料及進行漁場管理。本研究之目的在找出可區分和鑑定U. (P.) chinensis、U. (P.) edulis與U. (P.) duvaucelii三物種未成熟個體的形質特徵。
本研究先採用PCR-RFLP的方法解決未成熟標本的鑑定問題:先定序三物種成熟個體的16S rDNA片段,依據種間序列變異位找出脢切圖譜可區分三物種限制脢SspI。166隻無法經由形態形質確定物種的個體,先經PCR-RFLP處理鑑定後,確定為97隻U. (P.) chinensis、58隻U. (P.) edulis及11隻U. (P.) duvaucelii。物種確定後測量標本的16個形態形質,並將數據資料標準化成指數進行判別分析。結果顯示結合堅硬不易變形的幾丁質內殼及頸軟骨形態形質,即有83.7﹪的個體可被正確歸類。
雖然三物種未成熟個體的各形態形質存在嚴重的重疊現象,但經本研究結果可利用形態形質之判別函數分析或分子形質之限制脢RFLP方法,快速正確地區分和鑑別物種,藉此瞭解三經濟物種的生活史和孵育場選擇的問題,達到管理鎖管漁業資源之目的。
Uroteuthis (Photololigo) chinensis, U. (P.) edulis and U. (P.) duvaucelii are closely resemble in areas of sympatry. There is overlap in most diagnostic characters of the three Uroteuthis spp. Small specimens of three Uroteuthis spp. are more difficult to identify, and there are no studies detailing their morphmetry. Emphasis is placed on taxonomic characters suitable for use in the field.
Because immature specimens of the three Uroteuthis spp. are difficult to identify, the specimens need to be confirmed by PCR-RFLP (SspI), and morphometric character indices used in species separation are evaluated. Best discrimination between the three species are achieved with combination of nuchal cartilage length and gladius characters. The result shows 83.7% of the specimens can be correctly classified.
Although overlap is high for all indices in three Uroteuthis spp., we can use morphometry approach (discriminant analysis) and molecular biology technique (RFLP) to distinguish the three species. These information is useful for investigation and fishery management.
