老化的過程會伴隨著腦部與生理機能的衰退變化。本研究目的在探討給予核苷與核苷酸混合物(nucleosides-nucleotides mixture; NNM)、維生素E (vitamin E; Vit. E)及四氫生喋呤(tetrahydrobiopterin; BH4)對老化促進小鼠學習記憶能力與抗氧化能力之影響。實驗以7月齡老化促進小鼠(senescence accelerated mice;SAMP8)為實驗對象,分為控制組及飼料中添加0.5%NNM、0.01%Vit. E或0.001%BH4之實驗組,為期12週。實驗期間觀察記錄體重、攝食量及評估老化指數並測定動物行為活動量(open field activity test)、主動迴避試驗(active shuttle avoidance test)及單次被動迴避試驗(single-trial passive avoidance test)。動物犧牲後,血液方面檢測生化值、總抗氧化力、維生素E濃度;在組織方面檢測肝臟與腦部的重量及超氧歧化酶活性,並觀察腦部脂褐質及空泡數量。
實驗結果顯示各組間的體重、攝食量、活動量、血液生化值與組織重量均無顯著差異,而在老化指數、主動迴避試驗、體內維生素E濃度及腦中空泡化與脂褐質數量上,Vit. E與NNM的補充可以有效改善上述老化之現象(p<0.05)。體內的總抗氧化能力、腦部與肝臟的超氧歧化酶活性及維生素E濃度以Vit. E組的改善效果最為顯著(p<0.05);此外,NNM的給予對於體內抗氧化狀態也有提高之傾向。但添加BH4的飲食對於小鼠習學記憶能力、體內抗氧化防禦能力及腦部機能的幫助並不大。因此,飲食中添加NNM與維生素E,對於老化現象的進行有延緩改善之作用,而口服補充BH4則較無所助益。
The aging process accompanies pathological changes of brain and retardation of physiological function. The purpose of this study was to investigate the effects of 0.5% nucleosides-nucleotides mixture (NNM), 0.01% vitamin E (Vit. E) or 0.001% tetrahydrobiopterin (BH4) supplementation on learning and memory ability and antioxidative status in senescence-accelerated mouse prone-8 (SAMP8). Seven month-old SAMP8 used as subjects were divided into control group and experimental groups which were supplemented with NNM, Vit. E or BH4 for twelve weeks. Body weight, food intake, grading score, open field activity test, active shuttle avoidance test and single-trial passive avoidance test were performed during the experiment. After animals sacrificed, this experiment analyzed the biochemical parameters, total antioxidant states and vitamin E concentration of blood, weighed tissue weights, tested in superoxide dimutase activity on liver and brain of tissue and observed the brain histopathology of lipofuscin and spongy degeneration.
According to the results, body weight, food intake, open field activity test, blood biochemical parameters and tissue weight all had no significant difference between four groups. Vitamin E and NNM supplementation could improve the aging condition that deteriorated in grading score, active shuttle avoidance test, vitamin E concentration and brain histopathology (p<0.05). The improving effects of total antioxidant states, superoxide dimutase activity on tissues and vitamin E concentration were the most significant on Vit. E group (p<0.05). Furthermore, the antioxidative ability also was had enhanced tendency by the supply of NNM. But the BH4 supplement diet had less improvement for learn and memory ability and antioxidative defense ability and brain physiology. Therefore, the diets of supplementation NNM and Vit. E had the improving effect to delay the proceeding of the aging process, and oral supplement BH4 had less improving effect.
