目前已知動情激素具有促進細胞增生的功能,且可能涉及乳房組織和卵巢組織的致癌機轉,為了更完整的分析動情激素促進細胞增生的分子機制,我們利用microarray的技術分析加17β-estradiol處理後的OVCAR-3卵巢癌細胞株,總共挑到146個up-regulated genes及18個down-regulated genes,之後再從中選定一些基因利用RT-PCR與real-time PCR作進一步的驗證。
在結果中,本實驗找到兩個與DNA複製有關的基因,且這兩個基因還未被發現受動情激素調控;根據這兩個基因在5’端調節區可能都含有一段ERE的分析結果,推測這兩個基因可能是以ligand-dependent路徑受動情激素調控。而在進一步分析其他與DNA複製有關的基因後,發現這幾個基因的5’端調節區也都可能含有ERE。所以動情激素除了刺激cyclin D1表現而促進cell cycle進行外,還存在一條路徑是由動情激素依ligand-dependent方式,直接刺激細胞增生。
Estrogens induce cell proliferations and inappropriate responses of mitogenic actions occurred in the majority of malignant breast and ovarian tumor. However, the molecular mechanisms of estrogen signaling in cell proliferation have yet to be completely illustrated. In this study, we used the human OVCAR-3 ovarian cancer cell treated with 17β-estradiol to perform estrogen-responsive expression profiles using cDNA microarray analysis. After statistical analysis of the expressoin patterns, 146 up-regulated genes and 18 down-regulated genes were found. The expression pattern of several genes were confirmed by RT-PCR and real-time PCR.
Base on the results, two previously unreported genes related to DNA replication appeared to demonstrate estrogen-regulated expression. Analysis of the sequences immediately upstream of the transcription start site showed that they both have a putative ERE, so it is thought that the two genes are likely regulated by estrogen directly with the ligand-dependent pathway. Furthermore, the other genes related to DNA replication are also showed a putative ERE. Consequently, it could be concluded that in addition to cyclin D1 regulated by estrogen, there is another pathway of estrogen inducing cell proliferation.
