摘要
本論文的研究子題有二:(一)加速溶劑萃取-高效液相層析儀分析山質中多酚類活性成分之方法開發研究;(二)以液相層析-電灑式質譜儀分析植物性雌激素之研究。
研究子題一是以加速溶劑萃取山楂中多酚類等抗氧化的活性物質,加速溶劑萃取法利用加壓的方式提高萃取溶劑之沸點,另外,在惰性環境下萃取,可減少抗氧化物質在萃取過程中的氧化,並在提高萃取效率的同時,縮短中草藥分析前處理所需耗費的成本,有利於中草藥之科學化與品質管理。本方法的偵測極限介於0.5-7.5ng/g (乾重),定量極限介於2-25ng/g (乾重)。研究結果顯示,60%乙醇水溶液,在萃取條件:80℃、100bar進行循環萃取(3分鐘×3次),萃取回收率介於88-119%之間,每一個樣品萃取時間約為15分鐘,溶劑的使用量約減少至15mL。根據不同來源樣品定量分析的結果,以多變量統計分析,可以瞭解樣品受存放時間的影響。
研究子題二是以操作簡單的超音波振盪,進行中藥材與營養補充品中植物性雌激素萃取,並以液相層析串聯質譜儀進行定性、定量分析。製備空白樣品測試回收率結果介於94-100%之間。對於基質複雜的中藥材,串聯質譜儀的高選擇性,可以加強前處理上選擇性的不足。本方法的偵測極限介於0.01-0.2ng/g,定量極限介於0.1-0.5ng/g,方法重複性測試結果的相對標準偏差介於2.1-9.6%。定量分析結果顯示,單方中藥材飲片或科學中藥中的植物性雌激素以Daidzein、Genistein為主,其他營養補充品中以Formononetin較為常見。
Abstract
This thesis consists of two projects: (1) the determination of polyphenolics in Hawthorn by accelerated solvent extraction (ASE) and high performance liquid chromatograph- UV detection; and (2) the determination of phytoestrogens in Chinese herbal medicines and nutritious supplements by HPLC/ESI-MS.
The first project involves the application of accelerated solvent extraction method to extract antioxidants, i.e., polyphenolics in Hawthorn. This technology, based on increasing the boiling point of extracting solvents, is a solvent-less method operated in a closed system. It can prevent polyphenolics form degradation by light and air during the extraction stage. The analytical benefits will facilitate the modernization and quality control of herbal medicines. In the proposed method, the limit of detection (LOD) is 0.5-7-5ng/g (d.m.), and the limit of quantitation (LOQ) is 2-25ng/g (d.m.). Extracting with 60% ethanol solution at 80℃, 100bar in three cycles (3 min each), the recovery is from 88-119%. The quantitative results indicate that the contents of polyphenolics in commercial available Hawthorn are discrepant. The relation between the amount of polyphnolics and the storage time will be discussed by multivariate techniques.
The second project pertains to extract phytoestrogens in Traditional Chinese medicines (TCMs) and nutrient supplements by a simple extraction method, i.e., ultra-sonication, followed by LC/ESI-MS detection. In this method, the LOD is 0.01-0.2ng/g, and the LOQ is 0.1-0.5ng/g. the RSD of reproducibility (n=3) is 2.1-9.6%. The quantitative results reveal that Daidzein and Genistein are frequently detected in TCMs, and the main phytoestrogen in nutrient supplement is Formononetein.
