本研究利用四個microsatellite基因座的變異探討在台灣的青剛櫟族群之遺傳結構,再以遺傳結構去分析之前親緣地理之研究。此外,對於遺傳結構與地理空間結構的相關性也利用相關的檢測來檢定,分析是否青剛櫟族群的遺傳結構受到台灣地形結構的影響。
在所得結果中,四個microsatellite基因座的變異相當高(11 alleles/locus; HE = 0.741),但有顯著差異存在,四個基因座的FIS有兩個明顯大於0,另外兩個則是較接近於0,而如果族群採樣為隨機採樣的話,那造成此差異的原因可能是基因座上具有無效對偶基因存在。針對Mic69基因座所作的定序比對中,flanking region具有鹼基對substitution與deletion的變異,而後者會影響PCR產物的判定,而造成低估對偶基因的數目。
在台灣的青剛櫟族群明顯地偏離哈溫平衡(p<0.001),族群內明顯地有異型合子不足的情形,可能族群內近親交配頻繁,也可能有因族群合併而產生了Wahlund’s effect的影響,不過以基因座變化來看,或許可能為無效對偶基因的影響。而青剛櫟族群間則有分化不明顯的情形,FST最高為0.041,且從對偶基因頻率的分布與稀少的特有對偶基因產生,顯示青剛櫟的族群在台灣倒上有頻繁的基因交流,且地形的影響不大。
以microsatellite所得的之遺傳結構與之前青剛櫟親緣地理研究(Huang et al., 2002)有部份相符。兩者所得遺傳歧異度較高的三個族群(陽明山、霧社和浸水營)相同,而南部族群分化程度較高,也擁有較高的對偶基因豐富度,推測可能為 較靠近原始的族群,也就是冰期時的避難所。而霧社的高歧異度的原因可能是其位處台灣中部山區,在族群重新拓殖時,成為許多族群的交會點,而成為各族群遺傳組成的匯集點,因此具有高度的變異。從各族群間的平均分化值,發現扇平、浸水營與紅葉為分化值較高的三個族群,而從遺傳關係樹的關係中,發現扇平與浸水營為與其他族群分化程度較高的族群,顯示南部族群與其他族群的分化程度明顯,台灣南部可能為青剛櫟在冰期時可能的避難所。
